hello,
does anyone know what to use to dilute IBMX for cell culture. I want to
obtain the solution the more concentrate is possible. thanks in advance.

I have used IBMX, as a phosphodiesterase inhibitor, in cAMP accumulation
assays. The final concentration used was 500uM (micromolar) in the assay
medium (see Hirst et al., (1997) British Journal of Pharmacology 120,
509-515). This concentration was achieved by dissolving 5.5 mg of IBMX
(Sigma) in 200ul of DMSO and then adding this to 50ml of media.
Hope that this has been of some help.

Warren.

I usually prepare IBMX as 0.1M in DMSO and use it 1000x (100uM final).
Hope this helps

Hi Danielle,
IBMX is poorly soluble in aqueous solution. Dissolve a stock solution of
0.5 M in DMSO, aliquot and store at -20. For use, dilute to 1 mM (or
whatever concentration you use) directly into medium. It helps if the
medium you're adding to is stirring or lightly vortexing as you addthe
IBMX to avoid having it crash out of solution.
Steve

I've used DMSO. 10e-3 M stock diluted 1/100 in medium. Vehicle control
is very important.

Hi,

I dissolve IBMX in 50% ETOH to make my 10E-1 M stock solution and 1:1000
dilution in medium for assay. Sometimes U need to warm up (37°C, a couple
of minutes) the stock to get all IBMX into the solution.

Hope this helps,

Mohammad

Dear Warren, Based on Sigma, The dilution solution should be prepared in "Warm DMSO". But How Much warm. Thank you.
