G'day All,
I'm trying to transfect plasmid DNA into VERO cells, using a lipid DNA
complex. I've tried both Lipofectamine and Promega's new Tfx transfection
reagent. I've just done my first run through with a reporter plasmid to
make sure the system is working and have found the highest transformation
I got was 18 cells on one 6 well plate (pretty abysmal). I know the
transfections require optimisation of the amounts of Lipid to DNA and the
amount of time the reagent is applied to the media before addition of
complete media.
Cell death isn't a problem either, the vero's were quite happy
sitting there for 5 hours in a ml of reagent/medium mix (for teh
lipofectamine).
Has anyone out there transfected vero's using either lipofectin,
lipofectamine or the new Tfx reagent and consequently already done the
optimisation's i'm trying to do?

Any help would be greatly appreciated,
Yours sincerely, Alistair
