Query - Nucleic acid extraction from biofilms

Hi,

1. What is the most appropriate method for growing biofilms if the objective is to obtain atleast 2ug of DNA or RNA ?

2. I choose to grow them in static mode over a glass slide 1" x 3" immersed in a culture broth, will this method work?

3. If this be the method after how many hours of incubation will i be able to visualise the film layer ?

4. Alternatively can i use the method as shown in the following link for the aforesaid objective.

I need some expert advice on the subject.
