confocal microscopy related query

Dear Sir

 

i am working on a project on quorum sensing and quenching of pathogenic microbes.

 

Please help me with the protocol that i should follow to take a confocal scan of the biofilm.

so far i have checked the effect of a plant exact on the biofilm formation of pseudomonas. the treatment showed reduction in biofilm when quantified with crystal violet staining and 33% GAA washing method. further i wish to see the reduction effect by confocal microscopy. I donot wish to check the dead live cell counts. so can you please suggest how should i prepare the sample for confocal microscopy..can i use 35mm petri plate for growing the biofilm.
