Figures block like other obstacles, IIRC. 
So if you can get that tiny needed, you're good - but if you're tracing along the edge of their square, it's blocked.

You might consider a house rule of 50/50 miss chance when shooting around friendlies. Flip a coin - if it's heads, tails you miss, heads you *might* hit depending on the roll.

I just started playing Descent with my boyfriend and we love it! The only question I had was about Line of Sight. 
1) Can heroes attack through other heroes? IE: Can I have my hero friend duck so I can shoot an arrow? Vice Verse, can monsters/minions do the same?
2) If a hero and two minions are standing in a straight line of spaces, can the hero range attack the other minion (effectively shooting around the first minion) or is LoS blocked because the hero would have to shoot along a side of another figure?

So, I did check the manual. relevant passages are on pg. 8, 12, and 15 (emphasis mine):

pg.8:  "Figures cannot move into or through spaces containing figures or obstacles....These spaces are known as BLOCKED SPACES."
pg.12: "In order for a figure to have line of sight to a space, a player must be able to trace an uninterrupted, straight line from any corner of that figure's space to any corner of the target space. If the line passes through the edge of a map tile, a door, or a blocked space (a space containing a figure or obstacle), the target space is not in line of sight...."
p.15: "Knocked out heroes do not block line of sight." (This implies that heroes that are not knocked out do block line of sight).

So officially, it appears the answer to both your questions is no. To me, that's fair, and makes thematic sense. 

If that's unpalatable, I suppose you could house rule a "duck" action, or use the 50/50 suggestion from Shawn--but I'd say if you miss your 50/50, the attack should hit your ally...!

In reflecting some more (plenty of time now that BGG is down again), I suppose the house rule should really be an Awareness check for the ducking hero--and sure, if it fails, you still get to roll defense.

If it still bugs you that your teammates are in the way of your targets, then try using weapons/runes with blast!
