As my two munchkins continued to haphazardly fight over the up/down lever, I desperately tried to decipher the rhyme. Were we a red fish or a two fish? Were we supposed to go up up up or down down down?

Stellaris RNA FISH Probes hybridise to RNA. Genomic (double stranded) DNA remains inaccessible as no denaturation step is performed to unwind the DNA. Further, the Tm of the short, single-stranded probes is lower in DNA than in RNA and would therefore be competed away by the complementary strand. Simultaneous RNA and DNA detection using different probe types is not currently supported because the denaturation step needed to unwind DNA will remove the Stellaris RNA FISH probes.
Formalin-fixed paraffin embedded (FFPE) tissue may suffer from RNA degradation, sometimes very significantly. In such cases, detection of an RNA from a reference gene can be useful for verification of the experimental technique. One commonly used reference gene is glyceraldehyde 3-phosphate dehydrogenase (GAPDH). We make available several Stellaris ShipReady probe sets that can serve as controls, including human and mouse GAPDH, and DesignReady probe sets for many other organisms.
To best detect individual transcript molecules, we encourage the use of conventional wide-field fluorescence microscopes with a 60-100x, 1.3 NA or greater, oil-immersion objective and a cooled CCD camera. The light source should be a mercury or metal-halide lamp (e.g., ExFo Excite, Prior Lumen 200). We recommend starting with a 1 second exposure time. In addition, the degree of autofluorescence will depend on the fixation method used. Formaldehyde tends to crosslink fluorescent enzyme co-factors, such as flavins, whereas fixation with methanol/acetic acid tends to release them and wash them away from the sample. Prolonged fixation at elevated temperatures tends to exacerbate the autofluorescence. To assess the overall background fluorescence and to determine the contribution of non-specific probe binding, we recommend imaging a second sample in parallel that has not undergone probe hybridization - a no probe control.

For oligonucleotide suspension, we recommend preparing stock and working solutions using a TE buffer (10 mM Tris, 1 mM EDTA, pH 8.0) made with nuclease-free water. The EDTA serves to protect against microbial contamination. If your experiment cannot tolerate EDTA, you may use 10 mM Tris-Cl buffer. Suspension in water alone should be limited to nuclease-free water at physiological pHs, but is not recommended. Acidic conditions can lead to degradation of the oligonucleotide through depurination.
Notes: Fluorophores are sensitive to photobleaching. To minimize their exposure to light, we recommend using amber microtubes, wrapping the tube in foil, or else placing clear tubes into a box which is impermeable to light. Avoid freeze/thaw cycles by making working solutions and storing them in aliquots at -15 C or cooler.
We rate this attraction one out of five flying fish in terms of fear factor. This silly spinner ride is geared toward young children, and it may only be uncomfortable for those with a serious fear of heights or water (and we mean serious).
For the average parkgoer, this might be skipped unless small children are involved. Don't let that happen! While yes, this is just like Dumbo for all you Magic Kingdom people, it takes it to the next level for the slightly older kids with the challenge to not get wet. It's fun and I actually got pretty wet when I was on this ride (unintentionally). It is very basic so don't expect much, but it is tons of fun!
My four-year-old and I had so much fun on this ride. It is a simple concept, ride in a circle and control whether your fish goes up or down. Water shoots in a stream from fish on the periphery. My son thought it was the bee's knees getting wet while on the ride, but quickly changed his tune once we got off the ride and were soaked. So bring a change of clothes at least for little ones, ya dig?
My husband and I wanted to make sure to ride all the rides in the parks. We are in our late 20's with no kids and decided to get on One fish, Two fish...It was extremely cute. We didn't care if we were the only adults on the ride at the time. We would totally ride it again just because you pretty much get drenched whether you follow the rhyme or not.
We've been visiting Universal on and off since my son was 4. For those who think of that "other park" for little kids, they are missing some treats. The Seussland at IOA and the area at USO which includes "E.T.", Barney, and the ever fun Curious George area don't know what they are missing.
My son adored the whole Seuss area at IOA, but One Fish, Two Fish was by far his favorite. You can control your "fish" -- taking it up or down as you fly. There are water spouts which you can try to avoid or aim for, depending on your preference. The preference of a boy is to hit as many as possible!
There is music which livens up the mood, too. The ride is a lot of fun, whatever your age -- but we built up a lot of happy memories on that ride and it remains one of my favorites for the little ones. We preferred it to the other rides in Seussland -- and that is saying something as this is a colorful, fun filled area of IOA.
Think of the Dumbo ride at that other park... same concept, but MUCH beter.
Riders are counted and sent to the waiting platform (don't worry when you see parties of three seeming to cut the line in front of you- they're just going to the three fish cars!)
You get to choose your fish, and control it's up and down movements. Now the fun part... LISTEN to the rhyme and the directions in the song... it's quite adorable. It will say things like "Red fish down" and if you're in a red fish, you must go down unless you want to get squirted by a fish. SO FUN!!
LOVED the interactive element to this ride.. much more fun than simply up and down!

This works the other way around as well! If fish is started by something else, it inherits that parents exported variables. So if your terminal emulator starts fish, and it exports $LANG set to en_US.UTF-8, fish will receive that setting. And whatever started your terminal emulator also gave it some variables that it will then pass on unless it specifically decides not to. This is how fish usually receives the values for things like $LANG, $PATH and $TERM, without you having to specify them again.
