It's easy to get there by metro and bus.

You get to Berri-UQAM (the central hub of the metro) and take
the yellow line to Parc Jean-Drapeau (one stop). Metro map:
http://www.stm.info/English/metro/a-mapmet.htm

At Jean-Drapeau you change to bus 167. It's not very far, as
you'll see here:
http://www.stm.info/English/bus/PLAN_LIG/A-PL167.htm

(On this page the Metro Jean-Drapeau is given its old name
Ile Ste-Helene, but they are the same. This is the page for
the metro station with the first and last trains indicated:
http://www.stm.info/English/metro/a-m45.htm)

You might also ask at your hotel as I think there are various
private shuttles that make it even easier to go there. A taxi
to or from downtown would not be so expensive either - maybe
$12 or $15.

The Casino operates 24/7, but the metro only operates between
5:30 a.m. and 1:00 or 1:30 a.m.

Yes, you could drive, but enough people do drive that the
parking's a bit cramped at peak hours. Bus or taxi is the
way to go.
