Abstract
An important paradigm in evolutionary genetics is that of a delicate
balance between genetic variants that favorably boost host control of
infection but which may unfavorably increase susceptibility to
autoimmune disease. Here, we investigated whether patients with
psoriasis, a common immune-mediated disease of the skin, are enriched
for genetic variants that limit the ability of HIV-1 virus to
replicate after infection. We analyzed the HLA class I and class II
alleles of 1,727 Caucasian psoriasis cases and 3,581 controls and
found that psoriasis patients are significantly more likely than
controls to have gene variants that are protective against HIV-1
disease. This includes several HLA class I alleles associated with
HIV-1 control; amino acid residues at HLA-B positions 67, 70, and 97
that mediate HIV-1 peptide binding; and the deletion polymorphism
rs67384697 associated with high surface expression of HLA-C. We also
found that the compound genotype KIR3DS1 plus HLA-B Bw4-80I, which
respectively encode a natural killer cell activating receptor and its
putative ligand, significantly increased psoriasis susceptibility.
This compound genotype has also been associated with delay of
progression to AIDS. Together, our results suggest that genetic
variants that contribute to anti-viral immunity may predispose to the
development of psoriasis.

association between alcohol consumption and psoriasis has been
inconsistent among studies. To examine the magnitude of the risk of
developing psoriasis for drinking populations compared to those with
non-drinking, and to determine causes of the variation in odds ratios
(OR) between various case-control studies, we performed a
comprehensive published work search and a meta-analysis of case-
control studies considering prevalence. We did electronic searches on
Medline, and searched reports to identify case-control studies of
prevalent of psoriasis. We did meta-analyses of study-specific
incremental estimates to determine the risk of psoriasis associated
with drinking. The magnitude of the OR was analyzed by combining 15
case-control studies that matched defined criteria. The variance in OR
between studies was explored. The overall OR of psoriasis for drinking
persons compared to those with non-drinking was 1.531 (95% confidence
interval [CI] = 1.164-2.014, P = 0.002) and the association remains
statistically significant across a number of stratified analyses in
European descent subgroup (OR = 1.432, 95% CI = 1.085-1.889, P =
0.011) and also persists in sensitivity analyses performed to assess
the potential effect of varying psoriasis outcome definitions. Alcohol
consumption is associated with increased risk of psoriasis. These
epidemiological observations should inform the exploration of
biological mechanisms that link alcohol consumption with psoriasis.

This study investigated whether methotrexate, by interrupting the
methyl transfer function of folate, can induce genomic DNA
hypomethylation in patients with inflammatory arthritis. Consecutive
subjects with inflammatory arthritis (rheumatoid or psoriatic), who
were taking methotrexate (n = 7) or other medications (n = 6), and
control subjects, either healthy or with osteoarthritis and taking
nonsteroidal anti-inflammatory agents only (n = 9) were recruited. The
methylation status of genomic DNA from peripheral blood mononuclear
cells was determined. Plasma levels of folate, B12, and pyridoxal-5'-
phosphate (PLP), all of which are involved in biologic methylation,
were also examined. The extent of genomic DNA methylation was lowest
in subjects with inflammatory arthritis who were not taking
methotrexate, highest in subjects with inflammatory arthritis who were
taking methotrexate, and intermediate in control subjects (p < 0.05).
Plasma levels of folate and B12 were similar among the three groups.
The mean plasma PLP level in subjects with inflammatory arthritis was
33% lower than that in control subjects (p = 0.04). No significant
correlation between genomic DNA methylation and folate, B12, and PLP
levels was observed. These data do not support the hypothesis that
methotrexate induces genomic DNA hypomethylation. However, these data
indicate that inflammatory arthritis is associated with genomic DNA
hypomethylation that is reversed with methotrexate. Future studies
using a larger number of subjects are warranted to confirm these
findings.

Abstract
BACKGROUND AND AIMS:
A low dietary folate intake can cause genomic DNA hypomethylation and
may increase the risk of colorectal neoplasia. The hypothesis that
folic acid supplementation increases DNA methylation in leucocytes and
colorectal mucosa was tested in 31 patients with histologically
confirmed colorectal adenoma using a randomised, double blind, placebo
controlled, parallel design.
METHODS:
Subjects were randomised to receive either 400 microg/day folic acid
supplement (n = 15) or placebo (n = 16) for 10 weeks. Genomic DNA
methylation, serum and erythrocyte folate, and plasma homocysteine
concentrations were measured at baseline and post intervention.
RESULTS:
Folic acid supplementation increased serum and erythrocyte folate
concentrations by 81% (95% confidence interval (CI) 57-104%; p<0.001 v
placebo) and 57% (95% CI 40-74%; p<0.001 v placebo), respectively, and
decreased plasma homocysteine concentration by 12% (95% CI 4-20%; p =
0.01 v placebo). Folic acid supplementation resulted in increases in
DNA methylation of 31% (95% CI 16-47%; p = 0.05 v placebo) in
leucocytes and 25% (95% CI 11-39%; p = 0.09 v placebo) in colonic
mucosa.
CONCLUSIONS:
These results suggest that DNA hypomethylation can be reversed by
physiological intakes of folic acid.

Abstract
DNA methylation is one of the important mechanisms regulating gene
expression. Since beta-carotene (BC) was shown to have pro-chemotactic
activity and stimulates expression of pro-angiogenic genes, this study
was undertaken to define the possible changes in DNA methylation in
endothelial cell and its progenitors in the presence of BC. The
culture medium for human umbilical vein endothelial cells (HUVEC) and
endothelial progenitor cells (EPC) was supplemented with BC (1 - 10
microM) with the presence of arachidonic acid (AA) (3 microM). Global
DNA methylation tended to be lower in both endothelial cell lines,
after incubation with BC and AA. HUVEC incubated with AA demonstrated
the lowest DNA methylation. The decrease of DNA methylation in EPC,
induced by BC, was concentration-dependent. The microarray study
revealed, that the angiogenesis and homing-related genes were mostly
influenced by BC and AA in investigated cells. Our results indicate
that BC and AA-induced DNA hypomethylation in EPC and HUVEC, might be
a mechanism which may alter gene expression in endothelial cells what
in certain conditions may be connected with the suggested pro-
malignant effect of this compounds.
