Study: Drug used in treating cancer found to improve aging skin

Secondary benefits of prescription medications is not an uncommon find
in the world of medicine. In the past, patients treated with a
chemotherapy drug for certain cancers noted skin improvement. A new
study confirms anti-aging results.
A topical prescription cream currently on the market for treating
precancerous lesions caused by sun damage may have a secondary and
much more appealing use. A study published this month in the Archives
of Dermatology revealed that out of 21 patients on the study, nearly
90 per cent reported anywhere between mild to moderate improvement in
their skin's wrinkles and another 75 per cent stating sun damage was
much or moderately improved.
During the two-week study, patients between the ages of 56-85 who had
sun damaged skin applied the 5 per cent fluorouracil cream known as
Efudex twice daily to their skin. After some itchiness and facial
peeling, an improved dermal layer emerged, resulting in better looking
skin.
Some specific results include a decrease in the average number of
actinic keratoses lesions of 11.6 per person to 1.5 per person,
decrease and softening of wrinkles and improvements in dark skin
spots.
Official study conclusions explain the results as:
Topical fluorouracil causes epidermal injury, which stimulates wound
healing and dermal remodeling resulting in improved appearance. The
mechanism of topical fluorouracil in photoaged skin follows a
predictable wound healing pattern of events reminiscent of that seen
with laser treatment of photoaging.
The study lead, Dana L. Sachs, M.D., University of Michigan, stated in
a report that although the results were positive and the chemotherapy
drug was generally tolerated in it's topical form, there were some
minor problems with discomfort, with only one study patient
experiencing severe inflammation of the skin.
Despite the itchy peeling skin and the treatment being moderately
uncomfortable, 17 out of 19 happy users said they would pay for the
treatment directly out of pocket. The treatment would be less costly
than laser resurfacing, although the compared results of the two
treatments may not be exactly the same.
Patients who were treated with systemic fluorouracil for colon, head
and neck, pancreas and other cancers have noted a change in the
appearance of their skin, prompting the development of the cream. For
many, the future of better looking skin could be found in a tiny tube.

in supplement form can help halt UV-induced skin damage and skin
cancer.
Also, Frank Rizzo stated, "Because of the new studies and the
researcher's findings we have now formulated a new skin care product
using Polypodium leucotomas which we are planning to release later
this year called Pro Dermex for use in Dermatologists offices for
healthier skin and reduction of skin damage caused by the Sun. The
product will be actively marketed to the 16,000 practicing
Dermatologists in the U.S., with a focus on the 2,500 stocking

The constant exposure of the skin to oxidative stress results in
damage to cellular DNA and cell membrane lipids and proteins. To
combat this problem, the skin contains a number of antioxidants that
protect against oxidative injury. However, these cutaneous
antioxidants can be depleted by sun exposure and environmental
insults, resulting in an overload of oxidation products. Thus, topical
antioxidants that replenish the antioxidant capacity of the skin have
the potential to prevent oxidative damage. A number of natural
antioxidant ingredients also have anti-inflammatory properties, and
can be used in the treatment of oxidative damage such as photoaging
and perhaps even skin cancer. This article summarizes the active
components, pharmacologic properties, and clinical effectiveness of a
number of natural antioxidant ingredients including soy, feverfew,
mushroom extracts, teas, Coffea arabica (CoffeeBerry), Pinus pinaster

administered orally to mice inhibits ultraviolet B (UVB) radiation-
induced skin cancer formation. UVB-induced murine skin cancers occur,
in part, because of UVB-induced immunosuppression. Thus, we examined
whether PL inhibits UVB-suppression of the induction of contact
hypersensitivity (CHS) locally or systemically. METHODS: C57BL/6 mice
received standard drinking water or water-containing PL. In the local
model, mice were shaved on the dorsum and exposed to 3500 J/m(2) of
UVB radiation daily for 4 days. Control mice were not irradiated.
After the last irradiation they were sensitized to oxazolone topically
at the irradiated site. To examine the ability of PL to inhibit
systemic UVB-induced immunosuppression, mice were given 10,000 J/m(2)
of UVB radiation once and immunized at a non-exposed site 3 days
later. Six days after immunization (in both models), mice were
challenged on the ears with oxazolone and 24/48 h ear swelling
assessed. RESULTS: PL in drinking water significantly reduced the
inhibition of CHS observed with exposure to UVB radiation in both the
local and systemic models. CONCLUSIONS: The ability of PL to inhibit
UVB radiation-induced immune suppression may explain, in part, its
ability to inhibit UVR-induced skin cancer induction in mice.

The extracellular matrix (ECM) that gives tissue its structural
integrity is remodeled in skin aging/photoaging and cancer via the
increased expression/activities of matrixmetalloproteinases (MMP),
inhibition of the tissue inhibitors of matrix metalloproteinases
(TIMP), or inhibition of collagen synthesis. Transforming growth
factor-beta (TGF-beta), a predominant regulator of the ECM, is
inhibited in aging/photoaging and stimulated in carcinogenesis. P.
leucotomos (fern) extract has potential to counteract these
alterations via its antioxidant, anti-inflammatory and photoprotective
properties. The goal of this research was to determine the efficacy of
P. leucotomos to (a) directly inhibit MMP-1, 2, 3, and 9 activities,
(b) inhibit MMP-2, and stimulate TIMPs, fibrillar collagens and TGF-
beta in non-irradiated or ultraviolet (UV) radiated fibroblasts, and
(c) inhibit MMPs and TGF-beta, and stimulate TIMPs in melanoma cells.
To this purpose, we examined the direct effect of P. leucotomos (0-1%)
on MMPs' activities, and its effects on the expression (protein and/or
transcription levels) of (1) MMPs and TIMPs in dermal fibroblasts, and
melanoma cells, (2) TGF-beta in non-irradiated, UVA (2.5 J/cm(2)) or
UVB (2.5 mJ/cm(2)) irradiated fibroblasts, and melanoma cells, and (3)
types I, III, and V collagen in non-irradiated or UV irradiated
fibroblasts. P. leucotomos directly inhibited the activities of MMPs
as well as the expression of MMPs in fibroblasts, and melanoma cells
while stimulating the expression of TIMPs in these cells. P.
leucotomos stimulated types I, III, and V collagen in non-irradiated
fibroblasts, and types I and V collagen in UV radiated fibroblasts. P.
leucotomos had predominant stimulatory effects on TGF-beta expression
in non-irradiated or UV radiated fibroblasts, and inhibited TGF-beta
expression in melanoma cells. The effects of P. leucotomos were
largely similar to that of ascorbic acid. P. leucotomos demonstrated
dual protective effects on the ECM via its inhibition of the ECM
proteolytic enzymes and the stimulation of the structural ECM
collagens. The effects of P. leucotomos on fibroblasts and melanoma
cells may be partly via its cell-specific regulation of TGF-beta
expression and partly via its antioxidant property. The intake or
topical application of P. leucotomos may be beneficial to skin health,
in aging and cancer prevention or treatment.

Fernblock, an aqueous extract of the aerial parts of the fern
Polypodium leucotomos, used as raw material for topical and oral
photoprotective formulations, was fractioned by HPLC and the main
components with antioxidant capability were identified by means of UV
spectra, electrochemical detection, and MSn. Phenolic compounds were
identified as 3,4-dihydroxybenzoic acid, 4-hydroxybenzoic acid,
vanillic acid, caffeic acid, 4-hydroxycinnamic acid, 4-
hydroxycinnamoyl-quinic acid, ferulic acid, and five chlorogenic acid
isomers. Total ferric antioxidant capacity (FRAP) of HPLC eluted
fractions was measured. The results suggest that the herein identified
compounds support, at least partially, the antioxidant and radical
