Abstract
Psoriasis and atopic dermatitis (AD) are skin diseases that are
characterized by polarized CD4+ T cell responses. During the
polarization of naïve CD4+ T cells, DNA methylation plays an important
role in the regulation of gene transcription. In this study, we
profiled the genome-wide DNA methylation status of naïve CD4+ T cells
in patients with psoriasis or AD and healthy controls using a ChIP-seq
method. Only psoriasis patient T cells, not those of AD patients,
showed distinct hypomethylation (>4-fold) compared to healthy control
T cells in twenty-six regions of the genome ranging in size from 10 to
70kb. These regions were mostly pericentromeric on 10 different
chromosomes and incidentally coincided with various strong epigenomic
signals, such as histone modifications and transcription factor
binding sites, that had been observed in the ENCODE project implying
the potential epigenetic regulation in psoriasis development. The gene-
centric analysis indicated that the promoter regions of 121 genes on
the X chromosome had dramatically elevated methylation levels in
psoriasis patient T-cells compared to those from healthy controls (>4-
fold). Moreover, immune-related genes on the X chromosome had higher
hypermethylation than other genes (P=0.046). No such patterns were
observed with AD patient T cells. These findings imply that
methylation changes in naïve CD4+ T cells may affect CD4+ T cell
polarization, especially in the pathogenesis of psoriasis.

Abstract
Epigenetics is the study of differences in phenotype, in the absence
of variation in the genetic code. Epigenetics is relevant in the
pathogenesis of many skin diseases. In the case of the common skin
cancers, aberrant methylation of tumor suppressor gene promoters is
associated with their transcriptional inactivation. Environmental
carcinogens such as ultraviolet radiation and arsenic may act through
epigenetic mechanisms. Hypomethylation is associated with activation
of systemic autoimmune diseases, such as systemic lupus erythematosus,
subacute cutaneous lupus erythematosus and scleroderma. This may be
through a mechanism of immunological cross-reactivity with
hypomethylated DNA from pathogenic bacteria. Epigenetic factors may
also be relevant in the pathogenesis of psoriasis and other
inflammatory skin diseases, as well as in the pathogenesis of the
disorders of genomic imprinting with cutaneous features.

BACKGROUND:
DNA methylation is an epigenetic phenomenon that can modulate gene
function by up or downregulation of gene expression. Vitamin B12 and
folate pathways are involved in the production of S-
Adenosylmethionine, the universal methyl donor.

FINDINGS:
Brain vitamin B12 concentration and global DNA methylation was
determined in transcobalamin receptor (TCblR/CD320) knock out (KO)
(n=4) and control mice (n=4) at 20-24 weeks of age. Median [IQR] brain
vitamin B12 concentrations (pg/mg) in TCblR/CD320 KO mice compared
with control mice was 8.59 [0.52] vs 112.42 [33.12]; p<0.05. Global
DNA methylation levels in brain genomic DNA were lower in TCblR/CD320
KO compared with control mice (Median [IQR]: 0.31[0.16] % vs
0.55[0.15] %; p<0.05.).

CONCLUSIONS:
In TCblR/CD320 KO mice, brain vitamin B12 drops precipitously by as
much as 90% during a 20 week period. This decrease is associated with
a 40% decrease in global DNA methylation in the brain. Future research
will reveal whether the disruption in gene expression profiles due to
changes in DNA hypomethylation contribute to central nervous system
pathologies that are frequently seen in vitamin B12 deficiency.

Cure, I can't go as far as that claim. From the more than fifty people

How does it work?
The regimen I describe increases Vitamin B12 and Folic Acid and CoQ10

[...] HERE IS WHAT I DID TO END MY PSORIASIS

I used a two phase approach.

#1 I took vitamin B12 1000mcg sublingual liquid (B Total brand can be
ordered at 1 800 700 0770 $11.95)or outside U.S. at http://www.worlddrugstore.com
and Folic Acid 800mcg (any good brand, at drug stores, $3.00 100
pills )once a day for six weeks. Or until large amounts of flaking
comes loose. I used oil in a bath to help it along once it started.
Avoid scratching or picking, remember, let the skin heal. In two to
three weeks the areas of the skin should be lighter in color, flatter
and less flaking. Continue the daily B/Folic.

#2 To speed the healing, I then used a coal tar preparation (Brand
MG217 Ointment at drug stores $8.95) on the areas of the skin once a
day every night, agressively brush off flakes so they do not block the
absorption of the lotion. It's smelly so I shower it off in the
morning before going to work. This is also good because it allows the
skin to breathe and continue healing between applications. In aprox.
1.5 weeks the lesions begin to fade. I reduced the B12 and Folic Acid
to every other day and on alternate days I take Zinc, vitamin A and D
tablets to strengthen the skin and promote healing and continued with
the coal tar ointment. In three weeks from beginning with the lotion
the redness and flaking was reduced in area 75%. In a total of 4
months from the beginning only tiny bumps were still there and light
pink or bluish scaring remained but continued to fade. At this point I
began getting outside in shorts to get some sun, which also speeds
healing. In six months I was 99.9% clear. Recently I have been taking
CoQ10 100mg capsule each day. CoQ10 or sometimes called Coenzyme Q10
or NADH is a yellowish powder usually in capsule form, it is
manufactured by many different companies and is available nearly
everywhere vitamins are sold. My few remaining small spots shrunk to
gone in two weeks without any use of the MG217. I would begin day one
with the B12/Folic Acid and the CoQ10 in the dosages I mentioned each
day
This is not magic, it takes work and developing a regular routine not
missing a day of treatment. Important I continue (two years later)
with a maintenance regimen of 100mg CoQ10 daily and checking my self
at least twice a week for any tiny beginnings of new red marks. When I
find any I agressively remove the scale and treat them with the MG217
ointment and they clear to normal skin rather quickly.

This is the basic process.

Abstract
Interactions between adaptative and selective processes are
illustrated in the model of recursive causality as defined in Rupert
Riedl's systems theory of evolution. One of the main features of this
theory also termed as theory of evolving complexity is the centrality
of the notion of 'recursive' or 'feedback' causality - 'the idea that
every biological effect in living systems, in some way, feeds back to
its own cause'. Our hypothesis is that "recursive" or "feedback"
causality provides a model for explaining the consequences of
interacting genetic and epigenetic mechanisms which are known to play
a key role in development of cancer. Epigenetics includes any process
that alters gene activity without changes of the DNA sequence. The
most important epigenetic mechanisms are DNA-methylation and chromatin
remodeling. Hypomethylation of so-called oncogenes and
hypermethylation of tumor suppressor genes appear to be critical
determinants of cancer. Folic acid, vitamin B12 and other nutrients
influence the function of enzymes that participate in various
methylation processes by affecting the supply of methyl groups into a
variety of molecules which may be directly or indirectly associated
with cancerogenesis. We present an example from our own studies by
showing that vitamin D3 has the potential to de-methylate the
osteocalcin-promoter in MG63 osteosarcoma cells. Consequently, a
stimulation of osteocalcin synthesis can be observed. The above
mentioned enzymes also play a role in development and differentiation
of cells and organisms and thus illustrate the close association
between evolutionary and developmental mechanisms. This enabled new
ways to understand the interaction between the genome and environment
and may improve biomedical concepts including environmental health
aspects where epigenetic and genetic modifications are closely
associated. Recent observations showed that methylated nucleotides in
the gene promoter may serve as a target for solar UV-induced mutations
of the p53 tumor suppressor gene. This illustrates the close
interaction of genetic and epigenetic mechanisms in cancerogenesis
resulting from changes in transcriptional regulation and its
contribution to a phenotype at the micro- or macroevolutionary level.
Above-mentioned interactions of genetic and epigenetic mechanisms in
oncogenesis defy explanation by plain linear causality, things like
the continuing adaptability of complex systems. They can be explained
by the concept of recursive causality and has introduced molecular
biology into the realm of cognition science and systems theory: based
on the notion of so-called feedback- or recursive causality a model
for epigenetic mechanisms with relevance for oncology and biomedicine
is provided.

accumulation of bone minerals, including heredity, diet, physical
activity, gender, endocrine functions, and risk factors such as
alcohol, drug abuse, some pharmacological drugs or cigarette smoking.
The pathology of bone development during intrauterine life is a factor
for osteoporosis. Moreover, the placental transfer of nutrients plays
an important role in the building of bones of fetuses. The importance
of maternal calcium intake and vitamin D status are highlighted in
this review. Various environmental factors including nutrition state
or maternal stress may affect the epigenetic state of a number of
genes during fetal development of bones. Histone modifications as
histone hypomethylation, histone hypermethylation, hypoacetylation,
etc. are involved in chromatin remodeling, known to contribute to the
epigenetic landscape of chromosomes, and play roles in both fetal bone
development and osteoporosis. This review will give an overview of
epigenetic modulation of bone development and placental transfer of
nutrients. In addition, the data from animal and human studies support
the role of epigenetic modulation of calcium and vitamin D in the
pathogenesis of osteoporosis. We review the evidence suggesting that
various genes are involved in regulation of osteoclast formation and
differentiation by osteoblasts and stem cells. Epigenetic changes in
growth factors as well as cytokines play a rol in fetal bone
development. On balance, the data suggest that there is a link between
epigenetic changes in placental transfer of nutrients, including
calcium and vitamin D, abnormal intrauterine bone development and
pathogenesis of osteoporosis.

Abstract
DNA methylation is considered as a potential cause of aberrations in
regulation of gene expression during carcinogenesis. Therefore,
changes in DNA methylation patterns may be targets for
chemoprevention. In the present study, we investigated effects of all-
trans retinoic acid (ATRA), vitamin D(3), and resveratrol alone and in
combination with adenosine analogues: 2-chloro-2'-deoxyadenosine
(2CdA) and 9-beta-D-arabinosyl-2-fluoroadenine (F-ara-A), on
methylation and expression of retinoic acid receptor beta 2 (RAR beta
2) in MCF-7 and MDA-MB-231 breast cancer cell lines. Alterations in
methylation and expression levels after treatment of cells with the
tested compounds were evaluated by methylation-sensitive restriction
analysis (MSRA) and real-time PCR, respectively. RAR beta 2 promoter
in the tested fragment was partially methylated in MCF-7 cells and non-
methylated in MDA-MB-231 cells. In MCF-7 cells, all compounds, except
for resveratrol, inhibited promoter methylation and increased
expression of RAR beta 2. All natural compounds improved the action of
2CdA and F-ara-A on RAR beta 2 methylation and/or expression.
Combination of ATRA or vitamin D(3) with 2CdA was the most effective.
In MDA-MB-231 cells, only 2CdA, F-ara-A, and ATRA induced RAR beta 2
expression without any notable effects in combined treatment. Our
results demonstrate that both natural compounds and adenosine
analogues are able to reduce promoter methylation and/or induce
expression of RAR beta 2 in non-invasive MCF-7 cells. Furthermore, the
natural compounds improve effects of adenosine analogues, however only
at early non-invasive stages of carcinogenesis.

Abstract
All-trans retinoic acid (ATRA) induces cellular senescence via up-
regulation of p16 and p21; however, the action mechanism of ATRA is
unknown. Here, we show that ATRA induces promoter hypomethylation of
p16 and p21 via down-regulation of DNA methyltransferases 1, 3a, and
3b to facilitate binding of Ets1/2 to the p16 promoter and p53 to the
p21 promoter, resulting in up-regulation of their expression and
subsequent induction of cellular senescence in HepG2 cells. These
effects were mediated by retinoic acid receptor β₂ whose promoter was
also hypomethylated in the presence of ATRA. Therefore, ATRA can be
considered as an epi-drug in cancer therapy.
