Not sure if I get this right, but you want to respond with one message
to everyone starting a private chat to you? It's been a while since I
coded in mIRC, but this should still be within my grasp :)
on 1:TEXT:*:?:/msg $nick My Message
on 1 basically means everyone
TEXT means it looks for a text being sent to you
* is a wildcard. Basically means that it will respond on any text.
? restricts the command to just checking private chats
/msg $nick will send a message to the one sending you a private chat
My Message is the message you want to send.
Check the 'on text' part of the mRC help out for more details :)

The $nick will be exchanged with the nick used to send you a text
message.
In mIRC hit ALT-R, then select the 'remote' section. Fill it in there.
Using mIrc and leaving the PC, how can I send the same message,

So say you wanted to display the same message every minute until you
stop the timer?
/timer 0 60 /msg #channel <message>
Substitute the 60 with the amount of seconds you want in between the
messages (for instance, 600 would get you 10 minutes), the #channel
with the channel you want the message displayed in, and <message> with
the text you want to display.

If you type it in like that in any window, it'll just activate. If
connected to multiple servers, bear in mind to type the command on one
of the channels on the same server you want to display the message.
Otherwise it might message the same channel on a different server,
which can't be that helpful.
/timer off will shut it down.
If you want it to start every time you start mIRC, check the Tools ->
Options (reachable through the keyboard shortcut ALT-O ) under Connect
-> Options -> Perform
