Sure! Like absolutely NOT because of satic electricity,
which can build up in/on plexiglass. This can destroy
your motherboard, and any other electronics in there!
ZAP! And you pay for a new system.
Another problem is heat, plexiglass will store a LOT
of heat. Computers are happy COLD.

Both. But you'd be better off using glass, and aquarium
sealant, which is 100% clear silicone.

Also do not forget that Plexi will not stop EFI. I'm not sure I'd
feel comfortable using one for anything other than show.

As long as you remember to wear your tinfoil hat, everything should be
OK.
You might want to line your undershorts with foil too, just to be on
the safe side.

I wouldn't think that static electricity would be a big problem, since each
of the components will have its own chassis ground, and they'll all share a
ground through their cabling. As long as you avoid static discharge while
you're assembling it, I wouldn't think that plexiglass would cause any
unusual problems. I've seen pictures of working PC's built with plexiglass,
wood, milk crates, 8-track consoles, and other creative materials. Hopefully
some of the experts that lurk on this group can offer some insight.
However, I agree about the heat. Plastics have much lower heat conductivity
than metals (ever seen a plastic heatsink?). Be sure you've got plenty of
airflow through that heat-insulating case, and be sure that every
heat-generating component has a way to stay cool. They normally dissipate
their heat through their thermal connection to the steel/aluminum case.
The easiest solution might be to use a standard ATX configuration, but add a
second intake fan at the top. Put your hard drive(s) directly in the path of
the second fan, and put your CD drive(s) either above or below it, so they
can get at least a little bit of airflow. You might even want to put a
heatsink on some of the drives, if you can find a way to do it without
making them look cruddy. I think PCPowerCooling has a low-profile, 40mm
radial fan for applications like this. You should also definitely use a
PCI-slot exhaust blower. You'll need plenty of airflow across the entire
motherboard, since it won't be able to dump heat into the case through its
standoffs. The PCI cards ought to be ok as long as the motherboard stays
cool, since their main heat dissipation is through their PCI slot
connection. The video card will be ok if you rig up the slot blower so it
helps (rather than fights) the card's onboard blower... or if the card has a
passive heatsink, just put the slot blower right up next to it (I've had
good results cooling down my passive-heatsink Radeon this way).
Good luck finding an attractive way to hide fan filters in a plexiglass case
