Hi guys - I'm thinking of picking up the DNS-320L for using with ESXi over iSCSI.

Does the Alt-F iSCSI package have a GUI I can use to configure my iSCSI LUN etc?

No.
And iSCSI was not working on RC4/4.1?, don't know if it is working on RC5. The iscsitarget code maintainers didn't release nothing in the last years.
