1. Is iTunes (or any other program that uses sound) still running in your system tray? If so, whack it.
2. If you expand Advanced audio is Line In muted by chance?

    I am setting up a usb turntable on my Vista system to record LPs On
    the Sound window and the Playback tab shows the speaker Conexant High
    Definition Audio as working and default. The gray bars on the left
    fluctuate to green when I play something from iTunes and I hear it.
    The Sound window and the Playback tab also shows the USB Audio CODEC
    speaker working. With the turntable usb cable plugged in and a record
    playing the Recording tab on the Sound window shows the USB Audio
    CODEC Micorphone as working and default. The gray bars on the left
    fluctuate to green with a record playing but no sound comes from the
    speakers. None of the speakers on the Sound window and the Playback
    tab show any indication they are attempting to playback anything.
