Does it have to be png?

You can get an empty gif generated for a location using this

http://nginx.org/en/docs/http/ngx_http_empty_gif_module.html

That does not really achieve any more than having a 1px static file but it is convenient.

Whats your use case?
Not sure what your actual issue is.
