Hello! I'm wondering about subj. Didn't yet go deep in TW5 architecture, but remember than in single file version there was a significant slowing when tiddlers counts more then thounsand (it was several years ago).
Just wonder is there any "virtual DOM" like in React and VueJS libraries?

Yes,
Tiddlywiki uses it's own implementation of a virtual DOM. It specifically builds a widget tree, that then it's translated into a doom tree. Like react, any change to a node results on a re-render of all it's children, so there are some performance problems too.
