Tire squish

I've got a squish in my front tire. At first, I thought it was the dreaded spoke creak but it's not.  When removing the wheel and squeezing the tire I get the squish at one location. I am riding DT Swiss RR 440 mounted with René hurst 38 700c endurance tires. I know this is a wider tire than would be called for. I have always ridden wider tires than called for. I have tried deflating and reinflating tire multiple times To ensure proper seating of the tire to no avail. I am running it at 40 Lbs. 
Also when deflating to 20 lbs the squish appears at multiple locations along the rim not just one .
Help me fix this. 
What say the brain trust?

I had a similar squish noise 3 seasons ago… I was running Panaracer gravel kings and the tires were a couple seasons old mounted tubeless with Orange Seal. I discovered that the sealant had gotten down in between the layers of the tire and formed squishy bubbles.
I think the root cause might be a puncture that  the sealant sealed but it opened a pathway down into the layered construction of the tire.
I think you might want to dismount the tire and check (if you are running tubeless) turn the tire inside out and feel all around the inside  and see if you find the squishy spots…I am about positive that is the problem. My remedy was to replace the tire. 
I bet the Rene hurst tires and the gravel kings or most of the nice tires are all built kinda the same. Also, I am not using OrangeSeal anymore, not because I was dissatisfied with the product, but when I bought new Rene Hurst tires, I got their sealant too.

Oh yeah, too small of a tube can definitely do some odd stuff. I'm running too small of a tube right now on a bike, and it sounds awful. Also inflates weird. Ordered some proper sized tubes and will swap them out as soon as they arrive.

Please follow up and post what the remedy is. I still suspect that the inside layers might be delaminating but I guess an ill fitting tube is also a layer. 
It would be interesting to know.

Did you talc the inside of the tire and the tube ?  That helps things seat and eliminates any friction of the tubes, tire and rim. When I talc tires I sprinkle it inside the tire and use a small cloth to wipe it around and coat the entire interior. The tube I hold it in one hand and sprinkle the talc in hand and pull the tube though with the other, alternating sprinkling and pulling. Be generous, let it coat all the rubber. I've pretty much done this all my life.

You said that you made sure the tire was seated properly but sometimes it’s hard to tell. I’ve had a squish before even though I thought the tire was properly seated. My usual solution is to inflate past my desired pressure until I’m satisfied I’ve heard all the pinging to be had. Then I release pressure until it’s where I want it. It solved my swish problem.

Thanks for the advice.
A while back when I swapped wheels after a flat & I grabbed a 28-32 mm tube for that 38 mm tire.  
So I replaced tube with a 35-47 mm tube & after a short ride the problem appears to be solved.  Time will tell !
Btw running at 40 lbs.
