Hi all,

I've run PICRUST2 using the MetaCyc reference database on my bacterial 16S sequencing data.
When I looked at the output some pathways were related to methanogenesis (methanogenesis from acetate and METHANOGENESIS-PWY).
However, this pathway is only found in the Archaea Kingdom, right?
So, I'm wondering if anyone has any clues about what might have happened.
Before Picrust2 I removed all the sequences related to Archaea or unknown taxonomy.
Just to let you know, I used DADA2 for the sequence analysis.

Thanks :-)

Hey there,

You're right to question that pathway - it must be present due to a sufficient number of EC numbers in that pathway being called as present. This could be because some of the 16S sequences places poorly and may erroneously be clustered with reference archaeal sequences.

Cheers,

Gavin
