How Do Updates to the Portable Version Work?

I used MLO for Windows for many years and was used to updating the version whenever notified a new update was available. I just now got such a notification and was about to install it when I realized I'd need to point the update to my USB drive and suddenly remembered I'm using portable, which isn't mentioned in the update dialog.

When should I install the MLO for Windows updates?

I don't think it will update automatically. I'm a beta tester, so I've saved the link so I can download the zip file each time there's an update to the beta. I expect that you'll have to use the same process for the formally released version, every time there's an update to that notified: Download the zip file and then unzip the mlo.exe file and save it over the old version.
