I fiddled around with Mnemosyne quite a while, trying to let it show my multi-column card properly. No good at all. I seriously doubt it supports more than two sides at all.
I prepared a three-column wordlist (tab delimited, utf-8) txt, imported it to Mnemosyne which simply loaded it as two-side cards (first column on the front side, the second and third on the back side, or the vice versa for "sister card"). I did clicked the "vocabulary" mode or whatever. Are there any other options that I was supposed to check and did not? What is the matter?

Hi,
Make sure there are actually tabs, as opposed to spaces. Look at the attached example, it works.
Or perhaps the confusion stems from how Vocabulary cards work / are displayed in the browser.
