Spot detector

Hello!

I am trying to use spot detector on a piece of live imaging however it is failing to detect the spots I want.  I have played around with scale and sensitivity but am having no luck.

Any advice on how to accurately track my spots?!

ps. I eventually want to calculate the lifetime of these spots (they should not move during their lifetime) which I guess I will do by using spot tracking.

Hi Katie, 

why are you unsatisfied with your detections (too much, too few..). You can also attach an image so we can see whether we can find the right parameters.

Here is an image attached.  I am trying to track the progress of the brighter red spots seen in these images.  I believe that they are rather transient and most only appear in one image.  I would like to record how many frames each spot lasts for.

Dear Katie,

It seems that spot detector with scale 2 and threshold 30 do the job! I also ran the spot tracking with obtained detections and it also worked very well.

Thanks so much.  I saw Alexandre on Friday and he helped me with this.  We also used scale 2 but at a 50 threshold.
