C. elegans science project - help needed

Hi, 

I am doing a science project on how various compounds affect C. elegans inflicted with Parkinson's. 

I am using strain OW13 of C. elegans, in which aSynuclein (an indicator of Parkinson's) glows yellow. 

The compounds which I hope to test are in powdered forms. They are punicaligan, curcumin, and nicotinamide (all from Sigma).

Also, the strains arrive feeding on an E. coli lawn set in NGM (Nematode Growth Medium) Agar.

I am not sure how to insert the compounds into the diet of the worms. 

I currently have three ideas: 

1) Adding the powder directly onto the C. elegens/E. coli lawn.

2) Adding the powder to a culture of E. coli and feeding the C. elegans this diet. 

3) Adding the powder directly onto the NGM agar. 

Which method you think is best, or is there a better way to add these compounds to the C. elegans' diet?

I am sorry that I can't help you with the specifics of treating the C. elegans with your compounds but I can provide some advice on the wording you are using.  Be clear that what is being used is a model of Parkinson's disease in C. Elegans.  This is extremely important as what is learned in the model is not necessarily transferable to humans or other models.

Hi Paul,

when I was working with C.elegans (host-parasite interaction, infecting worms with pathogenic bacteria strains) I mixed the pathogens with E.coli or I made a lawn with pure pathogenic bacteria. I would mixed the powder with E.coli, so you can be sure, that C.elegans really feed the powder. The worms can avoid food or some regions with bad food/pathogens, but not, when there is no alternative or when it is a mixed food, so that they have no choice. I would control the feeding speed.  I would observe the feeding worm - E.coli and mixed food with powder - with the binocular. Most worms live on the NGM agar and are crawling  in the bacteria lawn, assumed there is enough food for many many worms.

I love the projects with C.elegans ..... ; )
