blind autopatching in vivo and continuous recordings

Hi Luke,

I am thinking about doing blind autopatching in vivo. There are lot of custom implementations out there and now I saw that now acq4 also has an autopatching module with Sensapex. I wonder if acq4 could be a general solution with other manipulators and pressure controllers, maybe it's already written in such a way?

The other thing related to this is continuous acquisition, or sweeps where both the start and the end is triggered (since we don't know in advance how long a behavioral trial will be). I assume the continuous recording would be easier to implement.. maybe it's already implemented?

Any advice is much appreciated.

Hi Marton!

I believe blind autopatch is already possible. If all goes well, I will be testing this in slices over the next few months. To turn this into a more streamlined operation will require some changes to the autopatch module--currently it operates from a set of known cell locations (for cultured cells), but I want to be able to just select a region of by slice and let the system blind-patch randomly or systematically within that area. Another lab may already have implemented something similar.. I have a little research to do there.

ACQ4 does support non-sensapex hardware (most of my lab uses scientifica hardware plus custom pressure controllers), and for blind autopatch you probably wouldn't need the accuracy provided by sensapex. Do you already have hardware for this, or are you trying to decide what to buy? I'd be happy to provide guidance in implementing support for whatever hardware you end up with; there isn't much documentation around that process.

Implementing continuous recording is possible, but would require some effort since everything else in the system is built around epochal recordings. It's been on my list for a long time but I never attempted it because I didn't have an immediate need for it. Again I'd be happy to provide guidance here if you want to change some code.
