Hi,

I was wondering how the observed - expected (O'-C') was calculated for a given bin-bin interaction in Juicebox. Based on the O'/C' value, which I assume to be (observed/average)/(control/average), I thought that O' - C' would be observed/average - control/average, but the numbers don't add up.

In the below example, O'/C' (44.609/40.026) = 1.115. However, 44.609 - 40.026 != 5.898. I have seen this for all the regions examined. Further, what exactly is the average value?

I am using Juicebox 1.6.11.

Thanks,
Adan

9:39,950,001-39,975,000

9:39,700,001-39,725,000
observed value = 68
expected value = 44.157
O/E = 1.54

control value = 42
observed/average = 44.609
control/average = 40.026
O'/C' = 1.115
O'-C' = 5.898

Hey Adan!

The average value for a given map region is the sum of all the contacts at the specified resolution divided by the number of bins at that resolution. As such, the observed and control maps have different averages.
For greater clarity, I will update the tooltip text to say the following (which we will release soon with the next update).
Please let me know if this makes sense.
