Sorry about no replying, we're all on really awful internet here.

Repairing permissions does seem to be one good solid way to make sure that the launchd stuff works.

I hit the big, repair disk permissions reset, reseting launch services  (turned the start at log in off and on)  It works fine again

After way too much time wasted trying to get it to work properly, com.growl.GrowlLauncher appears when launchctl list | grep growl is ran.

Run "launchctl list | grep growl" first before doing the simple instructions below so you can see it not appear and then appear after changing permissions on Growl.app bundle.

Fix: Highlight Growl.app in your applications folder. Click the '+' sign and add administrators. Give administrator Read & Write permissions. Go to growl general prefs. Switch start at login slider from off then BACK so it reads ON. Run the "launchctl list | grep growl" command again and there it is. You can probably revert permissions back to how they were after it writes to launchd, I have not tried it yet but I believe once it's written it will stick. Please let others know if this works for you so they can fix it as well.

Don't know much about launchd but thanks to those who advised checking with "launchctl list | grep growl"

@Chas4 - Did you follow my instructions changing permissions on growl.app then changing them back when your done? Or did you simply repair everything, reset launch services, and then toggled On/Off? I'd like to know so I can remove my change permissions fix, if it's really just a matter of once repairing/reseting toggling on and off to register the daemon... Thanks. Glad you got it working too. All about that toggle!

I did a reset launch services (via Onyx (was trying to fix something else))  and then restart, then repair disk permissions, (after I had noticed growl not load on start)  then toggled it on and off for good measure and it worked right since, 

only had the issue one  time it worked fine after the growl 1.4 update,  might be a bug somewhere, happened Sunday or Monday can't remember when it lost the auto start

I see, glad you got it working. As a note, my advice about adding admin to growl.app w/ read/write permissions makes no difference in launchd getting written. It is all about toggling to get it working. I had GrowlLauncher showing up in my launchctly list for a couple restarts but it has reverted to it's old ways. For some reason, maybe non-growl related, launchd seems to dump Growl from it's list but not any other app. I have been doing my maintenance like repairing permissions, reseting LS with Onyx as well. Oh well, launchd and growl are simply not going to work for me. My final fix has been to turn off start at login and use an applescript to launch Growl.app at login. If others have the same problems as me let me know and I'll share the applescript. Sucks, but at least it works.
