Hey all,
Anyone out there happen to have a culture of B.stearothermophilus that I
could get a sample of? I need to validate my autoclave, and I can't
afford the test strips or ampoules that would normally be used. The
local university's culture collection manager isn't allowed to give out
*any* strains, no matter how common or innocuous, for "IP Reasons". *Sigh*.

Sometimes tattoo parlors use little "spore strips" to test their
equipment.
I think the idea is that, after running them through their autoclave,
they send the strips off to a lab to get them tested.
Maybe you could buy/bum one from them and just test it yourself.
Of course the trick will be trying to find a tattoo parlor that
actually cares enough to test their equipment - good luck with that
one.

Actually, over here the odds are pretty good that they all test their
equipment; liability is taken pretty seriously in Ireland! We have a
word for it: "Compo-culture", the mindset of too many Irish people that
if they need money, they can always find an excuse to sue someone for a
contrived reason.

For example; if an employee turns up at work stoned out of his or her
mind and reeling drunk, and you dismiss them from work, you (the
employer) are liable for huge penalties for improper dismissal. You have
to give them two distinct warnings and suggestions for improvements to
their work ethic first.

So, thanks for that suggestion, it's an excellent idea! I'll start
ringing around tomorrow and we'll see what that turns up.

Tape works great, but among many other odd requirements of my EPA GMO
consent conditions is a requirement that I perform a monthly validation
on the autoclave/pressure cooker, specifically with live spores. So,
although I use autoclave tape every time, I still need stearothermophilis.

Update:
I went back into the university after having an epiphany. Their
ridiculous "IP" policy on strains probably didn't apply to strains as
part of disposable products. So, after asking around for some disposable
B.stearothermophilus ampoules or strips, someone kindly supplied me with
some ampoules.

So, I'm gonna put them in the ol' water-bath-kettle overnight and see if
anything grows! If something does grow, that's another culture
"Jailbroken" for community use; I'd happily share.
