HELP/DJ Studio Desktop woes

For the second time, the studio desk top would not play my audio clips, thus causing long gaps of silence and a frantic effort to repair the problem with limited success. All of this (for the second time in as many live broadcasts) AFTER I had cued all audio ready to play. I will detail here all the steps taken with the hope that someone out there can give me a clue as to what is wrong????
 
a. Thumbdrive containing audio clips of 14min, 15min,18min, 2min and 6min was inserted into DJ computer.
b. Began dragging audio from disc folder onto desktop
c. After dragging two (2) audio clips, a box appeared announcing that the desktop disc was full and that I needed to erase some selections before any more could be placed on desktop
d. I brought up iTunes AND Winamp and split the audio clips between both, which seemed to solve the problem.
e. Checked all audio clips and cued up iTunes to play first 14min clip.
f. Then, to heed the earlier warning, I removed all audio clips from the desktop into TRASH  (This, I realize may be the error)
g.When time came to play first audio clip.....NOTHING. A box appeared announcing that the computer "could not locate" the audio.
h. Attemped to play music clip....same thing, and another song....same thing. It "could not locate"
i. Played cd 3 music
j. took another thumbdrive and onto it loaded audio clips from my laptop again  then into DJ computer and then into iTunes. Voila! On the air.
k. When trying to play second audio clip which I had just loaded, the box again came up stating "could not locate"
l Attemped to play audio clips from Winamp and a different problem occured. The same problem that occured on my last show. When I pressed PLAY it would not play. A little red light (below the little green light) would flash for an instant.
I again had to reload onto winamp.
 
If anyone can take the time to figure out what's going on here, I would appreciate it.

Elliot,

Step f was definitely the culprit for your first failure. Starting
with step k it's hard to understand the cause of that failure.
