Title: Inner Ear Progenitor Cell Generation from Human Pluripotent Stem Cells
Abstract: Hearing loss is the most common disability in industrialized countries. Hearing loss is due to loss or dysfunction of the hair cells of the inner ear sensory epithelium. Hair cells cannot spontaneously regenerate in mammals, which leads to the permanency that typifies most cases of hearing loss. Mouse pluripotent stem cells can be used to generate functional hair cells in vitro however there are a paucity of reports of the use of human pluripotent stem cells for the same purpose. In this lecture we will discuss the generation of a population of inner ear (otic) - like progenitor cells from human pluripotent stem cells upon neural differentiation in vitro. Otic-like progenitors from alongside developing neural progenitor cells and express a panel of gene and protein markers indicative of an inner ear fate. Furthermore, the sequence of expression of otic markers occurs in a pattern that appears to recapitulate that seen during normal mammalian inner ear development. In addition, modifications in an established neural differentiation paradigm enrich this population of otic-like progenitor cells. Ongoing studies in our laboratory focusing on long-term differentiation of otic progenitor-like cells to a hair cell fate and inner ear transplantation will be discussed.
