Can TSE Pro be run as a singleton and subsequent invocation of it
(from command line) will load the file to be edited into the ring
buffer?

Yes.
Set Options, Full Configuration, System/File Options, Single Instance 'On'.

If you just want it set for a session (you do not want a permanent option):

g32 -1 somefile

To load another file into that instance:

g32 -1 anotherfile

Does this or should this apply to the console version (4.40a)? I can't
seem to get it to work, but I saw it in the help system.

The internal "single instance" code relies on features only present in
the Windows GUI API. However, there is a macro on our site that
performs a similar function, and I believe it also works for the
Console version.

The name of the macro is "onlyone".
