Abstract
Astragalus membranceus (Fish) Bunge Var. mongholicus (Bge) Hsiao is a
Chinese herb considered as an effective traditional anti-ageing
material. The two isomers of
4-hydroxy-5-hydroxymethyl-[1,3]dioxolan-2,6'-spirane-5',6',7',8'-tetrahydro-indolizine-3'-carbaldehyde
(HDTIC), HDTIC-1 and HDTIC-2, were extracted from the herb. We chose
them to investigate their effects on replicative senescence in vitro.

In this study, we observed the effects of HDTIC-1 and HDTIC-2 on
morphology, replicative lifespan, and specific markers related to
replicative senescence in human fetal lung diploid fibroblast (2BS
cell). Results have shown that both the HDTIC-1 and HDTIC-2 maintain
non-senescent phenotype of 2BS cells even at late population doubling
(PD) and increased cumulative population doublings (CPDs) by at least
15–20 PDs. The senescence-associated-galactosidase (SA--gal) positive
cell rates of late PD cells grown from early PD in medium containing
HDTIC, were much lower than that of late PD control cells, and similar
to that of young cells. HDTIC also improved cell growth and
proliferation and promoted the entry of 2BS cells from G0 or G1 phase
to S-phase. In addition, the advanced glycation end product (AGE)
levels of late PD cells grown from early PD in DMEM containing HDTIC
decreased significantly compared with those of late PD control cells.
Taken together, the results strongly suggest that both the HDTIC-1 and
HDTIC-2 delay replicative senescence of 2BS cells, and indicate that
the senescence-delaying effect of HDTIC appears to be due to its many
biological properties including its potentials of proliferation
improvement, inhibitory effect of AGE formation, and its antioxidant
activity. The differences of optimum concentrations of HDTIC-1 (0.1 M)
and HDTIC-2 (1.0 M) for delaying senescence also indicate that the
structure of HDTIC may be very sensitive to its activity.

Hi Kevin,
A quick search shows good in vitro studies of AR against
all the usual suspects, it seems this might very well be a good
addition to any regimin. No in vivo studies to speak of yet, but there
is a long Chinese tradition.
I have started a file with these papers representative of about
90, just aboaut all positive.

The previous experimental studies have demonstrated that addition
of Huang Qi ([symbol: see text] Radix Astragali) to the formulated
recipe Sheng Mai Yin ([symbol: see text] Decoction for
Pulse-activation) exerts the effects of strengthening the myodynamia,
increasing the coronary flow, improving myocardial metabolism, and
resisting the arrhythmia. The active component of Huang Lian ([symbol:
see text] Rhizoma Coptidis) can prolong the myocardial action
potential and antagonize the chloroform-, aconitine-, barium
chloride-, epinephrine- or coronary ligation-induced arrhythmia by

AIM: To investigate the inhibitory effect of a Chinese herb medicine
Astragali radix (AR) on growth of different cancer cell line. METHODS:
To observe the in vitro effects of AR on tumor cell proliferation by
trypan blue exclusion, MTS method and tritium thymidine incorporation
assay. Apoptosis was detected by DNA ladder method. RESULTS: The
inhibition rates of AR on the cell respiration of AGS, KATOIII, HT29,
MDA231, MEL7 and MEL14 were 68.25 %, 62.36 %, 22.8 %, 27.69 %, 2.85 %
and 5.14 % respectively at the concentration of 100 ug/ml; it
inhibited AGS DNA synthesis by 87.33 % at the concentration of 50
ug/ml. The inhibitory effect on AGS was time-and dose-dependent. AR
did not induce apoptosis in AGS cells. CONCLUSION: AR specifically
inhibits gastric cancer cells growth in vitro and the mechanism is
mainly cytostatic but not cytotoxic or inducing apoptosis.PMID:
12679907 FROM THE full text AR is a tonic herbal medicine which is
used as an adjuvant extensively in the treatment of various cancers,
such as lung cancer, digestive tract cancers, renal cancer, bladder
cancer[1,2,11], melanoma and AIDS[12]. It had been reported that it
could improve the host immune function suppressed by tumors and
relieve the marrow suppression induced by chemotherapeutic agents so
that it could make the patients more tolerant to chemotherapeutic
therapy. Although there were some studies showing that AR could
inhibit the tumor growth in vivo, the anti-cancer effect was testified
through activating the activity of Th, LAK and macrophages and
promoting the production of IL-2 and IFN-&#61543;. However, whether it
has direct inhibition effect on tumors cells has not been elucidated
yet.

Kevin..an EXCELLENT report that indeed follows the reputation of this
amazing herbal. When it is combined with Yerba Mate and Pau d'Aco(LaPacho)
an anti-aging phenomenon indeed occurs!, The addition of EYE spplements
