It may help to clear out any old mail he has on the server if it can be archived to a machine instead.
Make sure he has an active wifi or 3G connection before hitting send, its possible there is some connection negotiation that is happening in the background. Do speed tests to make sure the connection isn't the reason.

Take it to visit a genius at the store if he has Applecare on it.
Back it up in iTunes, erase & then restore the data he needs, perhaps setting up the mail account manually so that any old data isn't brought in from the iTunes backup.
It could just be that an update has started to play the sound when the server has confirmed the message was sent & they may have previously played the sound 1/2 way through the process. Mute his sound (Settings.app > Sounds) & see if he cheers up.
