Two chances: Fat and slim.
The chance is absolutely zero. Most of the OH 161/OH 37/OH 16/US 36
corridor that has been upgraded in Ohio west of Newcomerstown is at-
grade divided highway. There is no need to upgrade it to a full
freeway. If the segments along US 36 and US 250 east to Cadiz are
ever built, they'll probably be divided highways as well due to fairly
low traffic counts. (I don't see a tolled upgrade as ever happening
because it wouldn't offer a substantial improvement over the existing
roads.)
Beyond that, the states in question have long-standing history of
signing freeways as US and state routes. You're not going to see any
of them spend the money it would take on upgrades to bring the road up
to current Interstate standards.

I know a flyover ramp it probably cost-prohibitive for the area, but
did they consider revising the existing ramps and providing a short C/
D section on both sides? Just from a casual glance it looks like they
have enough ROW. (Disclaimer: not a CE.)

The Parkway will have a little C/D going on for that interchange and
adjacent one... but not Stubenville Pike (PA-60 & then 22/30)
I think looks may be deciving with the ROW. It's a pretty tight
interchange
Well, Giant Eagle @ that interchange has closed. If they can move the
Getgo, they would have more space.
