Running Defrag Prog

When I try to defrag my disks I get the following msg:

Disk Defragmenter: Defrag of (C:) has been aborted due to
inconsistancies that were detected in the file system.
Please run CHKDSK or SCANDISK on (C:) to repair. Then run
Disk Defrag.

When I run CHKDSK /v I get 'found error but could not
continue in read-only mode.

If I rin CHKDSK /f I get 'cannot lock current drive.
CHKDSK cannot run because volume is in use by another
program.

Any HELP will be appreciated. I am stumped.
