In case you mean that Vista, in Device Manager, doesn't detect the USB flash
drive, you then need to troubleshoot the issue. Here is a link that contains
steps to troubleshoot USB problems:

The site is for HP customers having USB issues, but these issues are
universal and can be used by anyone. Go through the list to determine
whether one or more of the steps can resolve your problem.

The solution to this annoying problem (I had it on one PC), seems to be to
locate and delete the file INFCACHE.1, which may be corrupted and causing
the problem. To delete this file, you will first have to modify its
permissions so the users group can access it (right-click on the
file->Properties->Security).
INFCACHE.1 file. It's located at C:\Windows\inf (scroll down to the file).
(Note: to see INFCACHE.1, you must set Windows Explorer to display hidden
and system files)
After deleting INFCACHE.1, I used Device Manager to remove my USB hubs so
Vista could rediscover them.

The drivers for this are built into Vista. We have seen that you may have to
force the computer to load the drivers. Try rebooting the computer with the
drive inserted. Also try having the computer look for the drivers on the C
drive and the Vista disk. Wait longer (<20 sec) to see if the driver
installation is slow on their system. During device driver search, try
pointing to this location.
Windows\System32\DriverStore\FileRepository
Enable "Include subfolders" in the Browse window.
If this fails, point to one of the subfolders within FileRepository:
FileRepository\usbstor.inf_...
FileRepository\disk.inf_...
FileRepository\volume.inf_...
FileRepository\wpdfs.inf_...
If FileRepository location(s) doesn't work, point to location.
Windows\System32\Spool\Drivers
Again, enable "Include subfolders" in the Browse window.
Check Device Manager for incorrectly installed drivers for the Data
Traveler. Uninstall them and reinsert the drive.
If the Data Traveler starts working ok, reinsert and make sure the drive
continues to work.

Finally the "resolution" for my DataTraveller is:
1. plug the device,
2. don't look for drivers (it will not find them);
3. delete the device from the Device Manager;
4. reboot the PC with the device in;
5. ensure that you see the new drive in "Computer" (wow! works!..);
6. when asked for driver (!) click "ask again later" or else the drive will
die (oops);
7. now you can use the flash drive as ususal, but each time when asked for
driver look up say "ask again later".
I see my DataTraveller twice in the Device Manager: in USB controllers and
in portable devices (translation from Russian, may be not the exact names).
If to press "Never ask again" when Vista asks to look for a driver, the
flash drive dies until the whole procedure repeated. :(
