I tried the sample tex file that came with the bundle (I am using
MacTeX2008) and the only error that I got was:

However, if I ignore this error (which seems to be some definition in
the cls file), then the document compiles OK. This makes me wonder
where you installed your bundle. If you installed it somewhere where
tex looks for sources etc, then you need to update your filename
database (for example, use texhash in linux). It appears that tex
cannot find some of the graphics. This will be a problem if the
directory structure has been changed form the one provided in the
bundle and the appropriate change has not been made in the source code
of the ample file.

To see if the sample file can compile at all on your system without
any changes, download a new instance of the bundle of files and unpack
on your desktop. Now move the pdf file that is already in the package
to somewhere else and try to compile the sample tex file. If this
works, it will mean that you need to install the package properly.

I do not understand the following lines of tufte-handout class :

Perhaps we are not speaking of the same thing. I have only one source
file: sample-handout.tex. What is the name of the source file you are
compiling? It is possible that I ended up downloading an older version
of the package. I'll try to get the newest version and then try it.

I just downloaded the package again and I see the same source file:
sample-handout.tex. On the ctan site:
