If a refund has been issued, but then a reversal of a refund is
required - is that possible through the API? Would putting a negative
number into Amount in SubmitOrderRefund do this?

No, the API does not support refund reversals. If you try to submit a negative amount, you will receive an error message stating that the refund request is invalid.
