Our Python / Boto program provisioned a 10,000-node cluster on Amazon
EC2 last month:

Boto works well even when the system has tens of thousands of objects
(esp. Spot Instance Request objects)!!

That's awesome!  Thanks for sharing.  It's wonderful to hear about how people are using boto.  This is a very cool application.
