Mating efficiency for pretransformed library screening 
  Hi zhenming: I find the "Mating efficiency for pretransformed library screening" in [link]. Now I have the same problem. I do the yeast 2-hybrid screen by mating system from clontech. The efficiency is very low. I use the Y2H gold and Y187. Do you have any advice to me? Thank you.... more »

SGD Servers Moving to New Data Center 
  Dear Colleague, This is a message alerting you of planned downtime for the Saccharomyces Genome Database resources for the week of November 27, 2012. Many SGD tools and resources will be unavailable from Tuesday afternoon, November 27th, until Thursday, November 29th because we will be moving our entire computing operation to a newly opened data center on Stanford campus. This new data center will provides a very robust environment including a more secure facility, a faster network, and a backup generator in the event of a power failure. There is also plenty of room for expansion. All of these features were missing from our current computing facility.... more »

S.cerevisiae GST expression vector 
  Dear Yeast community, We are looking for a S.cerevisiae expression vector that has a GST module, and also has a promoter and terminator, but not a carbon-source dependent promoter (eg. has a PGK or TEF promoter, rather than a GAL one). I haven't been able to find such a thing, but does anyone know if this exists and where we might be able to get it from?... more »

Help with IP and RNase A treatment 
  Dear all I'm doing co-Immunoprecipitation experiments in yeast (s. cerevisae) and I think the interaction I see between my two favorite proteins is bridge by mRNA (because both are able to bind RNA). I'm looking for a protocol to perform RNAse treatment of my extracts before IP. First I plan to go with RNAse A but someone advices me to not used it because it could not degrade poly(A)tail protected by the poly(A) binding protein and mRNA in the closed loop conformation. I'm very surprised because RNase A seems to be communly used for this application.... more »
