Isolation of pure bacterial culture from mixed culture

Please provide your input how can i proceed over the study of isolating the bacteria of my interest from mixed culture.
I know following steps:
1. serial dilution and plating on the diluted on the media plate
2. as per the morphological differences in cells separate the cells (I find all the cells similar not much difference) I label all cells
3. Next step is purify the colony by quadrant streaking (please explain over this part how can i proceed over this step?)
4. Screening the purified culture for the suitable biomolecule production.

Can any one suggest a more better method than the mentioned, any alternative?
