Novel Anti-inflammatory Molecules Of Microbial Origin

By studying the mycobacteria(1) that cause tuberculosis, molecules
with anti-inflammatory properties have been identified by the team led
by Valérie Quesniaux in the Laboratoire Immunologie et Embryologie
Moléculaires, working in collaboration with the Institut de Chimie
Organique et Analytique (both based in the University of Orléans/
CNRS). These molecules, as well as several new analogs, have been
synthesized. Their anti-inflammatory activity has been confirmed,
improved and demonstrated in the mouse, as reported in The Journal of
Biological Chemistry on August 28, 2009.

The mycobacteria that cause tuberculosis (Mycobacterium tuberculosis)
can remain silent for many years in man, thus eluding surveillance by
the immune system. In 90% of individuals, the infection is controlled
but not eradicated, and the mycobacterium can sometimes survive for
several decades. Mycobacterium tuberculosis produces several complex
proteins and glycolipids (a combination of sugars and lipids) that are
capable of modulating the immune response.

Valérie Quesniaux's team has focused on the interactions between the
mycobacterium and its host at the molecular scale, and on the
resulting immune mechanisms. On the cell wall of the mycobacteria, the
scientists identified low molecular mass glycolipids with very
interesting anti-inflammatory properties, the PIM (Phosphatidyl
Inositol Mannosides). They synthesized these PIM and reproduced their
anti-inflammatory properties. The team then turned to the synthesis of
novel, analog molecules, thus considerably simplifying their
synthesis. And indeed, the first example of this new family proved to
be even more active than the natural or synthetic molecules tested
hitherto. During their study, the researchers demonstrated the
enhanced anti-inflammatory activity of these novel PIM molecules both
in vitro and in an in vivo model of pulmonary inflammation.

Two patents have been filed in the context of this research(2) The use
of such immunomodulating molecules of microbial origin for therapeutic
purposes is seeing considerable growth. These new molecules could be
used in the treatment of severe, chronic, inflammatory diseases such
as rheumatoid arthritis, Crohn's disease or psoriasis.

Doug Melton: Finding a cure for diabetes

This week Doug Melton's team at the Harvard Stem Cell Institute in
Cambridge, Massachusetts, reported the first successful generation of
insulin-producing cells from the skin cells of people with type 1
diabetes. Roger Highfield spoke to Melton about the personal driving
force behind his research, and where he sees work on stem cells going
in the future.

Your son and daughter have diabetes, which must be a huge incentive to
develop new treatments for the disease. How far have you got?

My son Sam says to me: "Dad, this has taken you eight years and you
are telling me you have only solved two out of four or five steps.
What are you excited about?" I tell him that this shows it is do-able.
We will fill in the other steps.

In type 1 diabetes we have two problems. There is an absence of
insulin-producing beta cells and also an autoimmune attack, which is
what killed the cells in the first place. Our lab and others have
worked on problem one and I am confident that we are going to get
buckets of beta cells.

When I started working on this many years ago, I thought that by the
time I got to making beta cells the autoimmune problem would have been
solved by immunologists. In fact, it is a very nasty, difficult
problem.

If the autoimmune problem has not been solved, how are you going to
stop the implanted beta cells from being attacked?

The goal here is to find the root cause. I have that prejudice of
scientists – I need to know the mechanism behind the disease if I am
going to devise a treatment, rather than hope that something falls out
of the sky. The downside of this from my son's point of view is that
it will take a long time. But I see no reason I can't use induced
pluripotent stem cells (iPS cells) to reconstruct human diseases such
as diabetes, _____psoriasis_____ and multiple sclerosis.

If you wanted to design a thought experiment to get at the cause of
type 1 diabetes, it would go as follows. We would take a person with
diabetes and clone them 100 times. We would put the clones in
different environments, feed them different foods, give them different
viruses and every five minutes take blood samples, do pancreatectomies
and try to figure out the primary cause.

Obviously we are not going to do this experiment, but iPS cells give
us an alternative, which is why I am so excited. To investigate
diabetes, we need the immune system, since it is an autoimmune attack.
B cells, T cells, macrophages and dendritic cells – all of these come
from hematopoietic stem cells. Second thing we need is the thymus,
which is where your T cells go and are told what is self and what is
non-self. Thirdly, we need the target of the autoimmune attack, the
beta cells.

Our idea is to create these human cells from iPS cells and put them
into a humanised mouse, essentially a living test tube. The plan,
which will take me two years, will be to watch human diabetes develop
in a mouse. If that happens I would not even say it opens the door. It
opens a garage door, opens an airplane hanger, to a problem that no
one has been able to study. Then my first call is to work out which
cell screws up first.

Can you take this work further?

There's another aspect to this work which is science fiction. Just
imagine that I could make human thymic tissue in a dish to carry out T
cell education. In 30 years, you get a cancer, a melanoma. I say don't
worry Roger, I am going to take an iPS or a hematopoietic stem cell
from you and educate it with the thymic tissue to say that melanoma is
bad and should be killed. This is extremely powerful if it can be
done. That idea of educating T cells to kill melanoma has been done
quite effectively by David Baltimore at the California Institute of
Technology and his colleagues at the University of California, Los
Angeles.

Where do you see the future of stem cell research?

Fast forward with me. Why wait until you get sick? Why not use this to
keep us healthy? Stem cells are the key to understanding our natural
mechanism for repair and replenishment, which we could harness with
cells and drugs. I am not interested in immortality. I am interested
in living between the ages of 30 and 70 feeling more like a 30 year
old.

What do you think about the regulation of stem cells?

One nation, and only one, has a sensible approach to making policy on
this difficult subject, one that raises questions about what it means
to be human, when does life begin, what are the religious connections
and so on. I am a great fan of the way the British handled stem cell
policy. But the US is a mess. It is an embarrassment. Sometimes I
wonder how America can survive the way it makes decisions.

Sciences at Harvard University and co-director of the Harvard Stem
Cell Institute. He was studying frog development in 1993 when his
infant son was diagnosed with type 1 diabetes. His daughter Emma was

Cutaneous production of antimicrobial peptides (AMPs) is a primary
system for protection, and expression of some AMPs further increases
in response to microbial invasion. Cathelicidins are unique AMPs that

activity and (2) initiation of a host response resulting in cytokine
release, inflammation, angiogenesis, and reepithelialization.
Cathelicidin dysfunction emerges as a central factor in the
pathogenesis of several cutaneous diseases, including atopic
dermatitis, in which cathelicidin is suppressed; rosacea, in which
cathelicidin peptides are abnormally processed to forms that induce
inflammation; and psoriasis, in which cathelicidin peptide converts
self-DNA to a potent stimulus in an autoinflammatory cascade. Recent
work identified vitamin D3 as a major factor involved in the
regulation of cathelicidin. Therapies targeting control of
cathelicidin and other AMPs might provide new approaches in the
management of infectious and inflammatory skin diseases.

3. CLIMATE: OCEAN TEMPERATURES RISE TO A RECORD.
I still have global warming deniers sending me stories about how cold
it is
this summer in East Cupcake, Nebraska or someplace. But they didn't
send
me the Associated Press story this week on ocean temperatures. Because
of
its high specific heat, water temperature changes slowly. The
National
Climatic Data Center reported this week that the average global ocean
temperature in July was 62.6°F, the hottest since record-keeping began
in
1880. Another record: there are actually people in the water at
beaches in
Maine.

Small fluctuations in solar activity, large influence on the climate

Sun spot frequency has an unexpectedly strong influence on cloud
formation and precipitation

Our sun does not radiate evenly. The best known example of radiation
fluctuations is the famous 11-year cycle of sun spots. Nobody denies
its influence on the natural climate variability, but climate models
have, to-date, not been able to satisfactorily reconstruct its impact
on climate activity.

Researchers from the USA and from Germany have now, for the first
time, successfully simulated, in detail, the complex interaction
between solar radiation, atmosphere, and the ocean. As the scientific
journal Science reports in its latest issue, Gerald Meehl of the US-
National Center for Atmospheric Research (NCAR) and his team have been
able to calculate how the extremely small variations in radiation
brings about a comparatively significant change in the System
"Atmosphere-Ocean".

Katja Matthes of the GFZ German Research Centre for Geosciences, and
co-author of the study, states: „Taking into consideration the
complete radiation spectrum of the sun, the radiation intensity within
one sun spot cycle varies by just 0.1 per cent. Complex interplay
mechanisms in the stratosphere and the troposphere, however, create
measurable changes in the water temperature of the Pacific and in
precipitation".

Top Down - Bottom up

In order for such reinforcement to take place many small wheels have
to interdigitate. The initial process runs from the top downwards:
increased solar radiation leads to more ozone and higher temperatures
in the stratosphere. "The ultraviolet radiation share varies much more
strongly than the other shares in the spectrum, i.e. by five to eight
per cent, and that forms more ozone" explains Katja Matthes. As a
result, especially the tropical stratosphere becomes warmer, which in
turn leads to changed atmospheric circulation. Thus, the interrelated
typical precipitation patterns in the tropics are also displaced.

The second process takes place in the opposite way: the higher solar
activity leads to more evaporation in the cloud free areas. With the
trade winds the increased amounts of moisture are transported to the
equator, where they lead to stronger precipitation, lower water
temperatures in the East Pacific and reduced cloud formation, which in
turn allows for increased evaporation. Katja Matthes: "It is this
positive back coupling that strengthens the process". With this it is
possible to explain the respective measurements and observations on
the Earth's surface.

Professor Reinhard Huettl, Chairman of the Scientific Executive Board
of the GFZ (Helmholtz Association of German Research Centres) adds:
"The study is important for comprehending the natural climatic
variability, which - on different time scales - is significantly
influenced by the sun. In order to better understand the
anthropogenically induced climate change and to make more reliable
future climate scenarios, it is very important to understand the
underlying natural climatic variability. This investigation shows
again that we still have substantial research needs to understand the
climate system". Together with the Alfred Wegener-Institute for Polar
and Marine Research and the Senckenberg Research Institute and Natural
History Museum the GFZ is, therefore, organising a conference "Climate
in the System Earth" scheduled for 2./3. November 2009 in Berlin.

Greenpeace leader Gerd Leipold has been forced to admit that his
organization issued misleading and exaggerated information when it
claimed that Arctic ice would disappear completely by 2030, in a
crushing blow for the man-made global warming movement.

In an interview with the BBC’s Stephen Sackur on the “Hardtalk”
program, Leipold initially attempted to evade the question but was
ultimately forced to admit that Greenpeace had made a “mistake” when
it said Arctic ice would disappear completely in 20 years

The theory seems to hold water in the lab. In 2006, physicist Henrik
Svensmark of the Technical University of Denmark in Copenhagen and
colleagues produced aerosols artificially in an atmospheric chamber by
bombarding water molecules with a particle beam. "More ions resulted
in more aerosols," Svensmark says.

In the new study, Svensmark's team wanted to see if the idea also
worked in the real world. The researchers focused on a phenomenon
known as a Forbush decrease. Here, a massive storm on the sun's
surface flings a superhot fog of particles, called a coronal mass
ejection, past Earth, temporarily shielding our planet from cosmic
rays. If cosmic rays really do contribute to cloud formation,
Svensmark and colleagues hypothesized, then cloud cover should dip
during Forbush decreases.

And indeed that's what Svensmark's team found. When the researchers
examined cloud data collected by weather satellites over the past 22
years and compared them with 26 Forbush decreases, they discovered
that, for the five strongest events, the water-droplet content of
Earth's clouds decreased by an average of 7%. It's like bare patches
forming in a field, says Svensmark, whose team reports its findings
this month in Geophysical Research Letters. The cloud patterns
eventually returned to normal, he says, but they took weeks to do so.
"We're now convinced that aerosols are affected by the Forbush
decrease," Svensmark says.

Geoscientist Jón Egill Kristjánsson of the University of Oslo, Norway,
calls the findings "astonishing."

structures deep in the interior of the star, in a process analogous to
a sonogram in a medical office. "Using the global sound wave

between subtle changes in the Sun's interior and the sunspot cycle on
its surface," said Hill.

"This is an important piece of the solar activity puzzle," said Dr.
Dean Pesnell, of NASA's Goddard Space Flight Center. "It shows how
flows inside the Sun are related to the creation of solar activity and
how the timing of the solar cycle might be produced. None of the
forecasting research groups predicted the current long extended delay
in the new cycle. There is a lot more to learn in order to understand
how the Sun creates magnetic fields."

So many ask what do I do to get the aspartame out of my body, how do
I get rid of the symptoms and problems triggered by aspartame! Right

The most important starting point of any detoxification program is
to stop exposure to the toxin or toxins. This means avoiding even
small
amounts, since once sensitized to the toxin even minute amounts can
produce
full-blown toxicity. This is especially so with accumulative toxins,
such as aspartame.

It has been shown conclusively that the metabolic
products of methanol breakdown, formaldehyde in particular,
accumulates on the DNA and cellular proteins.

Once you have cleansed your diet of the toxin, removal of the toxin
and its metabolic products from your system will begin. Central to
this
process is the body's detoxification system,, which exist in all
cells of the body, with the bulk of detoxification taking place in the
liver.

The detoxification system is divided into two components called phase
I
and phase II, which work in tandem.

Toxins pass through phase I and then pass to phase II where they are
further detoxified and made water soluble for eventual disposal. It is
now known that you can significantly enhance the bodys ability to
detoxify these substances through the judicious use of specific
supplements.

Of the two systems phase II is most important and most often impaired.
The following is a list of nutrients that enhance detoxification:

* Indole-3 carbinol-100 mg twice a day. This natural substance
enhances phase II. It is extracted from cruciferous vegetables such
as broccoli, Brussel9s sprouts and cauliflower. It can be purchased as
an extract.

* Taurine 500 mg a day- This is a sulfur-containing amino acid that
is used by the liver for sulfonation reactions. It is to be taken
between meals.

* Milk Thistle (silymarin) 175 mg. Take two twice a day. This
extract has been shown to substantially enhance liver detoxification
and is
also a very powerful antioxidant for all cells.

* Curcumin- 500 mg three times a day with olive oil. This is an
extract taken from turmeric. It is an extremely powerful
antioxidant, enhances both phase I and II detoxification, is a
radioprotectant,
enhances bile flow, inhibits cancer growth, and is a very powerful
anti- inflammatory. Also enhances DNA repair enzymes.

* Panthethine 250 mg Take one capsule 3X a day- This vitamin is the
enzyme form of pantothenic acid . It plays a vital role in enhancing
the aldehyde dehydrogenase enzyme. (for formaldehyde detoxification)

* Thiamine- 100 mg. Take three a day. (vitamin B1) This vitamin
plays a major role in protecting nerves as well as the brain. It is
commonly depleted with pesticide exposure.

* Pyrodoxyl-5 phosphate 50 mg- Take one a day. This is the
functional form of vitamin B6. People exposed to chemical toxins
frequently have low levels of this vitamin.

When vitamin B6 is deficient, taurine is also low. It
plays a major role in protecting against glutamate and aspartate
toxicity.

In addition to detoxification, other nutrients play a more direct
role in protecting cells and in promoting their recovery from
toxicity.
These include:

* Magnesium- 750 to 1000 mg a day. (magnesium citrate/malate)
Magnesium plays a major role in cell protection, especially against
excitotoxicty. In addition it protects against strokes and heart
attacks.

* Selenium- 200 ug a day. This mineral also plays a role in
protecting cells from toxins, especially mercury. It plays a major
role in
protecting against free radicals.

* N-acetyl L-cysteine (NAC)- 750 mg a day. This is a substance that
in cells is converted to glutathione. Glutathione is your cell9s major
protection against free radicals produced by toxins such as
aspartame..

In addition, NAC removes mercury. NAC also enhances detoxicifation.
* Multi-3B2 vitamins- This should be a multivitamins that contains
25 to 50 mgs of each of the B vitamins. Take one a day.

* Vitamin E (as mixed tocopherols) 400 IU twice a day. A powerful
antioxidant, prevents heart attacks an stokes and the gamma-
tocopherol is a powerful anti-inflammatory.

* Vitamin C (as magnesium ascorbate) 2000 mg twice to three times a
day. Plays a major role in cell protection against oxygen stress,
protects the nervous system and enhances tissue repair.

* CoQ10 100 mg twice a day. This is a major energy molecule and
protects the brain against excitotoxicity.

* DHA 200 mg a day. This is a component of omega-3-fatty acids. It
plays a vital role in membrane repair and repair of synapses.

In addition to avoiding aspartame you must avoid other toxins as
well.

This includes MSG, pesticides, herbicides, bug sprays, fluoride,
mercury, cadmium, aluminum and the many other toxins found commonly in
the environment.

Many of these toxins act synergistically, that is, when
combined they have toxic effects that exceed their additive effects.

In addition, you need regular, moderate exercise (never aerobic),
avoid cheese, and refined vegetable oils.
Do not smoke or use drugs.

All of these things increase free radical damage and prevent tissue
repair.

You should eat a diet high in vegetables and with some fruits, no
sugar (or honey), moderate complex carbohydrates and pure water free
of contaminants, including fluoride.
Do not use fluoride tooth paste, mouthwashes or receive fluoride
treatments.

If you follow these principles you will notice quick recovery from
your toxic encounter. More serious toxic encounters may take several
months for recovery.

¦Explain that four lifestyle factors -- never smoking, BMI under 30,
exercise, and following a healthy diet -- were associated with almost
an 80% decreased risk of chronic diseases including diabetes, stroke,
myocardial infarction, and cancer.

Yet another study has confirmed that a healthy lifestyle can reduce
the risk of chronic disease, researchers say.
German patients who adopted four healthy lifestyle choices had a
nearly 80% reduced risk of major chronic illness including
cardiovascular disease, diabetes, and cancer, compared with those who
adopted none, according to Earl S. Ford, MD, MPH, a medical officer in
the U.S. Public Health Service at the CDC, and colleagues.

They reported their findings in the Aug. 10/24 issue of Archives of
Internal Medicine.

The researchers focused on four factors tied to20reduced disease risk:
never smoking, having a body mass index (BMI) lower than 30 ,
performing at least 3.5 hours per week of physical activity, and
following a healthy diet.

Aim: Tamoxifen is a nonsteroidal estrogen receptor modulator indicated
in the treatment of breast cancer. Apoptosis has been reported to be a
major mechanism for its antitumor effect. Tamoxifen has also shown
significant potential in treating various dermatological disorders
including psoriasis, characterized by hyperproliferation of epidermal
keratinocytes. An endeavor was made in the current studies to
investigate the potency of vesicle-encapsulated tamoxifen on human
epidermal keratinocyte cell lines. Methods: Drug was encapsulated in
the phospholipid-based vesicular systems, namely, conventional
liposomes and flexible-membrane liposomes. In vitro cytotoxicity
evaluation of the formulations was carried out employing MTT cell
proliferation assay. Results: A composition-dependent strong
inhibition in the viability of epidermal keratinocyte cells was
observed. Conclusion: The encouraging findings of this work construe
immense potential of the tamoxifen-encapsulated vesicular systems in
the management of psoriasis.
