Celiac.com 04/12/2010 - A team of researchers recently set out to look
at connections between psoriasis, the liver, and the gastrointestinal
tract.

The team was made up of Paolo Gisondi, Micol Del Giglio, Alessandra
Cozzi & Giampiero Girolomoni. They are associated with the Section of
Dermatology and Venereology of the Department of Medicine, at the
University of Verona, Italy.

Psoriasis is a common chronic inflammatory, immune-mediated skin
disease that is often tied to other disorders, including psoriatic
arthropathy, chronic inflammatory bowel diseases, and cardio-metabolic
disorders.

Additionally, about 50% of all patients patients with psoriasis suffer
from non-alcoholic fatty liver disease, from 0.2–4.3% suffer from
celiac disease, and about one half of one percent suffer from Crohn's
disease.

These associated conditions may have some common genetic traits, as
well as common inflammatory pathways, and their presence offers
important implications in the global approach to treating psoriasis.

In particular, common systemic antipsoriatic drugs might have a
negative affect on associated cardio-metabolic conditions and
nonalcoholic fatty liver disease, and may have important interactions
with drugs commonly used to treat psoriasis.

Moreover, the team emphasizes the importance of encouraging psoriasis
patients to drastically improve their modifiable cardiovascular and
liver risk factors, especially obesity, alcohol and smoking intake,
because improvements could have positive impact on both the psoriasis
and the patient's general well-being.

A Tumor Suppressor In The Gastrointestinal Tract

Recql5 is a member of the mammalian RecQ helicase family. Genetic
mutations that result in loss of RecQ helicase activity give rise to
disorders that are associated with cancer predisposition and premature
aging, such as Bloom, Werner, and Rothmund-Thomson syndromes. This
class of enzymes is best known for their role in maintaining the
integrity of the genome to prevent oncogenic mutations.

A research article published in the World Journal of Gastroenterology
addresses this question. A research group at Case Western Reserve
University examined whether this helicase actually has a role in tumor
suppression in the GI tract but this could not be assessed by a simple
study of straight Recql5 knockout mice because of the potential
inherent limitation of modeling GI tumorigenesis in mice. This group
therefore conducted a study by introducing the Recql5 mutation into
Apcmin mice, an established model for assessing the impact of various
factors on tumorigenesis in the GI tract. They found that indeed,
Recql5 mutation had an important impact on tumorigenesis in the GI
tract.

This new finding therefore has identified Recql5 as a tumor suppressor
in the mouse GI tract. Moreover, since mouse Recql5 and its human
counterpart are highly conserved, these findings have also raised the
possibility that RECQL5 may also have a role in human GI malignancies.
Moreover, Recql5 expression was recently shown to be an important
determinant in sensitivity to camptothecins, a class of anticancer
drugs that are currently used to treat human colon cancer patients.
Thus, RECQL5 may be an important candidate for a colon cancer
biomarker.

The fundamental knowledge regarding the molecular basis of oncogenesis
provides invaluable information that is vital for the identification
and development of novel biomarkers, drug targets or more effective
treatments for cancer patients. In particular, dysfunction in DNA
damage response and repair is a common feature in many human cancers,
including colon cancers. Significantly, this feature of cancer could
potentially be exploited for the development of novel diagnoses and/or
treatments.

The Rewiring Of Gene Regulation Across 300 Million Years Of Evolution

As published in Science, researchers from Cambridge, Glasgow and
Greece have discovered a remarkable amount of plasticity in how
transcription factors, the proteins that bind to DNA to control the
activation of genes, maintain their function over large evolutionary
distances.

The text books tell us that transcription factors recognise the genes
that they regulate by binding to short, sequence-specific lengths of
DNA upstream or downstream of their target genes. It was widely
assumed that, like the sequences of the genes themselves, these
transcription factor binding sites would be highly conserved
throughout evolution. However, this turns out not to be the case in
mammals.

The authors traced the evolution of gene regulation by comparing the
binding of evolutionarily conserved transcription factors in the
genomes of five vertebrate species - human, dog, mouse, short-tailed
opossum and chicken - spanning 300 million years.

In all tested species, the transcription factors CEBPA and HNF4A are
master regulators of liver-specific genes. By mapping the binding of
CEBPA and HNF4A in the genomes of each species and comparing those
maps, they found that in most cases neither the site nor the sequence
of the transcription factor binding sites is conserved, yet despite
this, these transcription factors still manage to regulate the largely
conserved gene expression and function of liver tissue.

Austrian Discover Lymphocytic Growth Factor

Austrian scientists have found the "gene switch" of leucopoiesis,
according to a report published by the professional magazine of Nature
Immunology.

A research group led by Wilfried Ellmeier, an expert from the Center
of Pathophysiology, Epidemiology and Immunology of the Medical
University of Vienna, discovered the important function of a factor to
CD8-positive T-Lymphocytes formation, in the process of the formation
of Gen MAZR.

T cells, which belong to the cell mass of leukocyte and play a very
important role in immunologic system, are divided into two main
groups, CD4-positive "Helper"-T-Cells and CD8-positive cytotoxic T-
cells, respectively.

The two groups of cells come from the Thymus consisting of precursor
cells which are also called CD4 and CD8 double-positive (DP)
Thymocytes. CD8 cells can destroy the attacked cells while CD4 ones
play an additional role as regulators in the process.

Ellmeier and his research group have explained the formation mechanism
of the immunological cell successfully, proving that transcription
factor MAZR participates in the immunological cells as well as in the
formation process of precursor cells, double-positive (DP) Thymocytes,
CD4-positive cells and CD8 positive T cells.

The research team have proved in their former studies that MAZR can
regulate the formation attributes of the superficial structure of CD8
cells.

Their latest research findings have confirmed the resolution that a
gene called "Th-pok" play a determinative role in the formation
process of CD8 positive cells that are regulated by MAZR.

If MAZR is impeded, precursor cells result in auxiliary cell of CD4.
Contrarily, CD8-positive cytotoxic T-cells are formed

Abstract
The CD4 versus CD8 lineage specification of thymocytes is linked to
coreceptor expression. The transcription factor MAZR has been
identified as an important regulator of Cd8 expression. Here we show
that variegated CD8 expression by loss of Cd8 enhancers was reverted
in MAZR-deficient mice, which confirms that MAZR negatively regulates
the Cd8 loci during the transition to the double-positive (DP) stage.
Moreover, loss of MAZR led to partial redirection of major
histocompatibility complex (MHC) class I-restricted thymocytes into
CD4(+) helper-like T cells, which correlated with derepression of Th-
POK, a central transcription factor for helper-lineage development.
MAZR bound the silencer of the gene encoding Th-POK, which indicated
direct regulation of this locus by MAZR. Thus, MAZR is part of the
transcription factor network that regulates the CD8 lineage
differentiation of DP thymocytes.

Molecular analysis of the zinc finger factor MAZR

Cell fate specifications during thymocyte differentiation are linked
with the tightly controlled expression of CD4 and CD8 coreceptor
molecules. The expression of the adjacent genes Cd8a and Cd8b is
controlled by at least five developmental stage-, subset- and lineage-
specific cis- regulatory elements. Our laboratory recently identified
with yeast one-hybrid screens the BTB/POZ-domain containing zinc
finger protein MAZR (Myc-associated Zn finger related factor) as an E8
II core enhancer binding protein. MAZR ( encoded by the Zfp278 gene)
is highly expressed in DN thymocytes and its expression is down-
modulated with progressive thymocyte differentiation. We could show
that Cd8 enhancers recruit MAZR to the Cd8ab gene complex and that
MAZR interacts with corepressor N-CoR (nuclear receptor corepressor)
complexes in DN thymocytes. Enforced expression of MAZR during T cell
development unexpectedly resulted in variegated expression of CD8 in
DP thymocytes, suggesting that MAZR negatively regulates chromatin
modifications at the Cd8 gene loci required for the activation of CD8
expression. In my PhD project, I am performing gain and loss of
function approaches to further analyze the role of MAZR.

Abstract
In recent years, the complexity of the 5' region of the Vitamin D
receptor (VDR) gene has become apparent. Six exons, 1a-1f, lie
upstream of the translation start codon in exon 2. Transcription has
been reported beginning in exons 1f, 1a, 1d and 1c and alternative
splicing can produce a large number of alternative mRNA transcripts.
Exon 1d transcripts can code for two alternative proteins. This
pattern of transcription produces several potential promoter regions.
We have used a number of in silico tools to study the evolutionary
conservation of the exon and promoter sequences of the 5' region.
Those exons involved in the alternative VDR proteins, exons 1d and 1c,
were well conserved from mouse and rat, unlike exons 1f, 1e and 1b
which showed little homology. Exon 1a was also well conserved.
Furthermore, 1a was shown to be within a strong CpG island and TraFac
revealed a Sp1-MazR-Sp1-MazR cluster of transcription factor binding
sites immediately upstream of exon 1a, a common motif in strong
promoters. The promoter region upstream of 1f showed a highly
conserved pattern of transcription factor binding sites and was also
shown to be within a CpG island. No significant clusters of conserved
sites were observed in the 1c promoter region, despite reports of 1c
promoter activity.

Abstract
Autism spectrum disorders (ASD) and attention-deficit hyperactivity
disorder (ADHD) are common and complex neurodevelopmental conditions.
Diagnostic criteria for these conditions have traditionally relied
solely on behavioral criteria without consideration for potential
biomedical underpinnings. Newer evidence, however, reveals that ASDs
are associated with: oxidative stress; decreased methylation capacity;
limited production of glutathione; mitochondrial dysfunction;
intestinal dysbiosis; increased toxic metal burden; immune
dysregulation, characterized by a unique inflammatory bowel disease
and immune activation of neuroglial cells; and ongoing brain
hypoperfusion. Many of these same problems are common features in
children with ADHD. These medical conditions, whether co-morbidities
or etiopathogenic, would be expected to have synergistically negative
effects on the development, cognition, focus, and attention of
affected children. It is likely these biological abnormalities
contribute significantly to the behavioral symptoms intrinsic in these
diagnoses. However, treatment for these underlying medical disorders
is clinically justified, even if no clear immediate behavioral
improvements are observed. This article reviews the medical literature
and discusses the authors clinical experience using various biomarkers
for measuring oxidative stress, methylation capacity and
transsulfuration, immune function, gastrointestinal problems, and
toxic metal burden. These biomarkers provide useful guides for
selection, efficacy, and sufficiency of biomedical interventions. The
use of these biomarkers is of great importance in young children with
ADHD or individuals of any age with ASD, because typically they cannot
adequately communicate regarding their symptoms.

classification of psoriasis as a systemic disease that exhibits a host
of co-morbidities. As a consequence, the second Interdisciplinary
Conference on Co-morbidities and Lifestyle Modification, convened by
the International Psoriasis Council, has concluded that specialist
physicians, primary care physicians and dermatologists are faced with
an opportunity to impact, not just psoriasis disease understanding and
management, but overall patient well-being. The conference panel was
represented by the disciplines of dermatology, cardiology,
rheumatology, epidemiology, endocrinology, hepatology and
gastroenterology, and medical specialists with particular expertise in
obesity, diabetes mellitus, inflammation and genetics. The multiple co-
morbidities associated with psoriasis were reviewed with a view to
identify possible mechanisms linking psoriatic disease with obesity,
metabolic syndrome, diabetes, cardiovascular disease and non-alcoholic
fatty liver disease. Consensus was established on the association of
psoriasis with other co-morbidities and disease states. Consequently,
there is a significant opportunity for specialist and primary care
physicians to collaborate with dermatologists in the management of the
overall health of psoriasis patients. First, there is an important
need for physicians to routinely screen psoriasis patients for the
multiple susceptibility risk factors and co-morbidities associated
with psoriasis. Second, the design and implementation of lifestyle
modification plans including exercise, diet and the limitation of
alcohol and tobacco intake, will not only benefit their general
medical health but also their psoriasis.

The U.S. Supreme Court will be holding a private conference hearing on
Friday, April 23rd 2010, to determine whether to hold a full court
hearing on corruption charges within the Los Angeles County government
for illegally imprisoning attorney Richard Fine. A rally to
acknowledge the U.S. Supreme Court hearing will be held at the L.A.
Superior Court building at 8 am Tuesday April 20th, by supporters of
Richard Fine’s release from illegal imprisonment. Mainstream media has
avoided any mention of this epic corruption scandal, despite its
significance, and now it is on its way to the the Supreme Court.

Richard Fine, who holds a PhD in international law and served as an
anti-trust prosecutor at the Department of Justice in Washington D.C.,
has been in jail in the L.A. County Jail for over a year in solitary
confinement. He never had a trial, there has been no conviction, nor
any sentence to keep him there. Sheriff LeRoy Baca claims he does not
know why Fine is in jail, yet he keeps him there and failed to answer
Fine’s Writ of Habeas Corpus. Baca refused to allow a filmed interview
with Fine until Judicial Watch filed a lawsuit on behalf of Full
Disclosure Network, and then, out of the blue, he “changed his mind”
and granted a filmed interview with them!

Fine believes he is imprisoned because of political reasons:

1. Fine exposed L.A. Superior Court judges of taking illegal money
from the County, in violation of the California State Constitution.

2. The County Supervisors were using public tax money to pay off the
Superior Court judges.

3. The Superior Court Judges decided cases in favor of the County
(Fine reports that since 2005 to present, only 3 people have won cases
against the County as a result of the bribes).

4. The California State Legislature have given the judges and County
Supervisors immunity from prosecution for their actions and
misappropriation of taxpayer money.

5. The judges have tried to get Mr. Fine disbarred from the California
State Bar as a result of the lawsuit he filed on behalf of the
taxpayers who were denied “due process”.

6. Superior Court Judge David Yaffe (accused judge) failed to recuse
himself and sent Fine to prison. He also failed to cite any precedent
(denying Fine “due process”).

7. L.A. County Sheriff LeRoy Baca illegally imprisoned Fine, and Fine
is in jail today.

8. Fine should have been released within 5 days, according to the law.
