Thankyou for your message.I will only be able to contemplate going back on the AMN 107, if Novartis allows me to take a much-reduced dose, as the 400 mg is much too toxic, and I have had to stop taking it until I go bak to Newcastle next Wednesday. Keep in touch and I will let you know what happens.

My husband is on Gleevec and doing well, I am a friend of Ruth's. AMN107
trials are soon to start in Singapore where we live and our doctor was
asking if any AMN107 patients I know have suffered adverse side-effects.
Would you mind sharing your side-effects with us?

Is anyone out there on a smaller dose than 400mg? I have had to stop the drug for the time being, as it seems to be too toxic for me. My consultant is away till next week, so I had no-one to ask for advice.
Feeling very despondent about it. Anyone else had a similar experience?

[AMN107] New Clinical Trials Now Recruiting For VX-680 For T315I Mutation

Clinical trials are now recruiting patients for the new Merck drug VX-680 which has been developed to work on the T315I mutation. There are 3 separate trials that are now open with trial locations in Texas, Massachusetts, New Jersey, Pennsylvania, and Wisconsin. CML patients who have failed on Gleevec, AMN107, and BMS354825 may be eligible for this trial. This is a potentially a very important drug for CML patients because it may work on the T315I mutation. If you know of any CML patients with the T315I mutation let them know about these trial locations so can find out if they may be eligible.

AMN107 is a small molecule tyrosine kinase inhibitor developed, in the first instance, as a potent inhibitor of BCR-ABL. We tested its effectiveness against fusion tyrosine kinases TEL-PDGFRbeta and FIP1L1-PDGFRalpha, which cause chronic myelomonocytic leukemia and hypereosinophilic syndrome, respectively. In vitro, AMN107 inhibited proliferation of Ba/F3 cells transformed by both TEL-PDGFRbeta and FIP1L1-PDGFRalpha with IC50 values < 25 nM, and inhibited phosphorylation of the fusion kinases and their downstream signaling targets.

I was diagnosed with CML on 2/28/04 after being hospitalized due to severe left shoulder and arm pain which the doc thought may be the beginning of a heart attack. My WBC was 82000 on Thursday and an oncologist was consulted. We live in a small town and I had just retired from a surgery office. One of my "surgeons" came by and talked with me and gave me more advise and info that any of the other docs had. By then it was 5PM on Friday and I was being discharged. My surgeon called a cancer clinic about 65 miles away and had me an appointment for Tuesday, March 2, 2004 and CML was confirmed by bone marrow. On that day my WBC was 135000.
